This research concerns on the best extraction process of the betalain assisted by microwave. The betalain was purified by means of macro absorption resin. The body oxidant resistance of its waterextract and the purified betalain was compared. Then the activity of the body anti-oxidant and anti-fatigue of betalain was studied. This study can serve as a reference for the further development and utilization of betalain. The results were showed as follows:(1) Response surface methodology determines the microwave extraction technique. This study reached the order of the factors that influence the extraction rate of the betalain. The order were:microwave time> microwave power>(vb:ma). The best processing conditions are: (vb:ma) is 47.2:1, microwave power 367.8W, microwave time 94.2s. The extraction rate of betalain was 18.41%in this case.(2) In order to improve the purity of anthocyanin, we employed ethanol precipitation, gradient polar solvent extraction and purification of macroporous resin successively. After comparison, it was proved that X-5 macroporous resin was the best fixed phase of the purified betalain. Static adsorption dynamics experiment showed that the absorption equilibrium can be achieved around 180min, and the best elution was ehanol of 60%. Dynamics experiment showed that the absorption effect was the best when the dynamic sample flow rate was 1 mL/min, and the elution effect was the best when the elution flow rate was 0.5 mL/min and the elution volume was 4BV. In this case, the betalain colourity was 61.18.(3) After the in vitro antioxidant activity experiments of crude extract of betalain and the purified betalain and their comparison with Vc, it was showed that they have remarkable scavenging effect against·OH, DPPH·and O2-·,showed response relationship with concentration, haved certain reducing power and significant anti-lipo peroxidation effect against lipid and grease, and that the antioxidant activity of the purified batalain was enhanced.(4) It also studied the function of betalain to the anti-fatigue ability of mice and the anti-oxidant ability of the mice after swimming. The result showed that betalain can prolong the forced-swimming time of mice, decrease the amount of BLA and BUN, accelerate their clearance speed, increase the amount and recovering rate of MG and LG and it has anti-fatigue effect. This study also showed that after exercise, the T-AOC of the serum and liver of small rat is improved, the SOD and GSH-PX enhanced and the amount of MDA reduced, thus proving the anti-oxidant function of betalain.
