Boletus edulis Bull:.Fr, is a renowned wild edible and medicinal fungi which hasmedicinal valve of antioxidant, anti-fatigue, anti-inflammatory and anti-tumor activity.For further study on the development and utilization of Boletus edulis resources, wedetermined the content of main immunomodulatory activity components mannanoligosaccharides and dextran of polysaccharides in Boletus edulis fruiting bodies by HPLCtechnique. This article tested Boletus edulis main ingredient of anti-tumor activity, studed thegrowth inhibition effect to tumor cells and screened the optimal anti-tumor concentration ofpolysaccharides and abstract of different polarity in vitro experiments. Then we explored theanti-tumor effect of Boletus edulis pharmacological on tumor-bearing mice with H22and westudied the effect of Boletus edulis polysaccharides on immune function of normal mice. Thepurpose is to provide a theoretical basis for Boletus edulis pharmacological research and drugdevelopment.1. We determined the main immunomodulatory activity components by HPLC techniquein Boletus edulis polysaccharides, the concentration of mannan oligosaccharides is11%anddextran is17.6%.2. This article tested the anti-tumor effect of Boletus edulis polysaccharides and75%alcohol extract,95%alcohol extrac, petroleum ether layer, acetate layer and butanol extractionlayer to SKOV-3human ovarian cancer cells and human breast cancer MCF-7cells in vitro byMTT. We provided different concentrations of administration, and calculated the inhibition rateof cells. The inhibition rates of Boletus edulis on ovarian cancer SKOV-3, the highest inhibitionrates of polysaccharide in400(μg/mL) was22.57%; the highest inhibition rates of75%alcoholextraction layer in400(μg/mL) was43.67%; the highest inhibition rates of95%alcohol extractlayer in10(μg/mL) was31.08%, the inhibition rate of petroleum ether extract layer in50(μg/mL) was39.92%; the inhibition rate of ethyl acetate layer was31.02%in400(μg/mL);inhibition rate of butanol extract layer in10(μg/mL) was16.13%. The inhibition rates ofBoletus edulis on MCF-7cells, the highest inhibition rates of polysaccharide in400(μg/mL)was19.12%, the inhibition rate of75%alcohol extract layers was30.09%in100(μg/mL); theinhibition rate of95%alcohol extraction in10(μg/mL) was20.38%; the inhibition rates ofpetroleum ether extraction in400(μg/mL) was29.84%; the inhibition rates of ethyl acetatelayer in50(μg/mL) was29.20%. 3. In the experiments of the anti-tumor effect and immune function of Boletus edulispolysaccharides on H22tumor-bearing mice, Boletus edulis polysaccharides can enhance theindex of each dose group of mice immune organ, high-dose group (BEBP1,500mg/kg), middledose group (BEBP2,250mg/kg), low-dose group (BEBP3,125mg/kg) for H22inhibition rateswere29.17%,31.60%,40.02%, of which the low-dose group of polysaccharides was thehighest inhibition rate of40.02percent, followed by the low-dose group were Boletus edulisdose group and high dose group. Low-dose group can significantly increase levels of IFN-γ,CRP, IL-6and TNF-α in tumor-bearing mice body, and polysaccharides in the other groups canimprove the body’s IFN-γ, IL-6, TNF-α levels, high-dose group can reduce the levels of CRP.4. Study the effects of Boletus edulis polysaccharides on immune function in normal mice.Firstly, Boletus edulis polysaccharides can make the weight of normal mouse improvesignificantly, and improve the thymus, spleen and liver organ index, and it also can increaseIFN-γ, TNF-α levels in the serum of the mouse and lower serum CRP, IL-6levels. Thequantitative PCR showed Boletus edulis polysaccharides can improve the gene of IL-8R inmRNA expression levels of the samples mouse’s intestinal, and reduced IL-6, TNF-α, CRP inmRNA levels.
