Objective:To study the positive expression of VEGF,p53 and CerbB-2 in colon cancer tissue,analyze their relationships and the correlation with hepatic metastasis and lung metastasis in colon cancer,and to find out objective parameters for prediction of hepatic metastasis and lung metastasis in colon cancer.Methods:100 patients with colon cancer that clinical and pathological data were complete,were underwent surgical resection between 2002,1 to 2009,1 at the First Affiliated Hospital of Jinan university.We test the expressions of VEGF,p53 and CerbB-2 in the tissue of 100 colon cancer(experimental group) and 20 normal tissue by the side of colon cancer(control group) with immunohistochemical technique(SP),explore the correlation between the expressions of VEGF,p53 and CerbB-2 and metastasis in colon cancer.Studying the correlation in simple expression of single protein and combined expressions of three proteins,to the hepatic metastasis and lung metastasis in colon cancer.Analyze the expressions of VEGF,p53 and CerbB-2 proteins are influential factor to the hepatic metastasis and lung metastasis in colon cancer.Results:1.The expressions of VEGF,p53 and CerbB-2 in 100 colon cancer tissue:the positive rate of VEGF,p53 and CerbB-2 are respectively 62%(62/100),74%(74/100), 36%(36/100).In 20 normal tissues by the side of colon cancer are respectively 20% (4/20),10%(2/20),5%(1/20).There is significant difference between two groups.2.The single expression of VEGF,p53 and CerbB-2 protein expression in hepatic metastasis and lung metastasis of colon cancer.The positive percentages of VEGF in hepatic and lung metastasis were 51.6%,25.8%,The levels were significant higher than the other sides(7.9%,5.3%).The positive percentages of p53 in hepatic and lung metastasis were 45.9%,24.3%,The levels were significant higher than the other sides(3.8%,0%).The positive percentages of CerbB-2 in hepatic and lung metastasis were 72.2%,33.3%,The levels were significant higher than the other sides(14.1%, 9.4%).There is significant difference between two groups(p<0.05).3.The combined expressions of VEGF,p53 and CerbB-2 protein expression in hepatic and lung metastasis of colon cancer,which were positively correlated with hepatic and lung metastasis in colon cancer.The combined expressions were easier having metastasis than the single expression of them.There is significant difference between two groups(p<0.05).4.The spearman analysis indicates that there is a correlation among VEGF,p53 and CerbB-2 in colon cancer;There is positive correlation between the combined expressions of VEGF,p53 and CerbB-2 and hepatic metastasis and lung metastasis in colon cancer. Furthermore,there is more correlative in combined expressions of three proteins than that of two proteins than that of single protein.Conclusion:Colon cancer with positive expression of VEGF,p53 and CerbB-2 proteins are easier to promote invading and happen metastasis;VEGF,p53 and CerbB-2 proteins play an important role in the mechanism of colon cancer invasion and metastasis. They can be used as parameters for the invasion and metastasis of colon cancer in molecular biological level.And they have some reference value to predict the depth of invasion of colon carcinoma,metastasis and prognosis.
