Research On The Effects Of BuShenHuaYuJieDu Recipe To The Lung Cancer Metastasis And The VEGF And MMP-9 Gene Expression In Lung Cancer

Objective: To observe the effect of Chinese herbal compound Bushenhuayujiedu recipe on mice with Lewis lung carcinoma, to explore its mechanism and to provide scientific evidence for the clinical application of Bushenhuayujiedu formula on the treatment of lung carcinoma in transferred and advanced stage.Methods: Mouse models of spontaneous metastasis of Lewis lung carcinoma were set up by implanting Lewis lung carcinoma cell and Bushenhuayujiedu formula were given to treat them. Weight of tumors, spleen and thymus in mice were measured. Carcinoma metastases in lung were observed. Expression of vascular endothelial growth factor (VEGF) protein and matrix metalloproteinase-9(MMP-9) protein were detected by immunohistochemistry. VEGFmRNA expressions was assayed by RT-PCR.Results: Bushenhuayujiedu formula had an obvious inhibited effect on implanted Lewis carcinoma and lung metastasis in mice. (P <0.05), and the formula can also promote the inhibition rate of Western medicine crisplatin(DDP) on the above two carcinomas. Bushenhuayujiedu formula can significantly improve the mice's immune function, particularly for the high-dose group (P <0.01). The expression of VEGFmRNA, VEGF protein and MMP-9 protein were decreased in Bushenhuayujiedu formula. Compared to simple DDP group and model group, the expressions of the above index in the Bushenhuayujiedu formula high-dose group were dramatically decreased (P <0.01).Conclusion: Bushenhuayujiedu formula obviously inhibited the implanted Lewis lung carcinoma and its spontaneous metastasis;Bushenhuayujiedu formula improved immune function of mice with Lewis lung carcinoma, which contribute to inhibiting the implanting cancer and the metastasis; Bushenhuayujiedu formula significantly reduced protein and the gene expression related to metastasis,and it can promote the therapeutic effect of the chemotherapeutics DDP on the disease.
