Objective: To study the positive expression of vascular endothelial growth factor VEGF,Cyclooxygenase-2 COX-2,Matrix metalloproteinase-9 MMP-9 in colon carcinoma tissue, analyze their relationships and the correlation between clinical and pathological data. To explore the role of the combined expressions of VEGF,COX-2,MMP-9 in colon carcinoma cells of joint invasion and lymphatic metastasis, and to find out objective parameters for prediction of colon carcinoma.Method: 50 patients with colon carcinoma that clinical and pathological data were complete were underwent surgical resection between 2000,1 to 2007,9 at the First Affiliated Hospital of Jinan university. We test the expressions of VEGF,COX-2,MMP-9 in the tissue of 50 colon carcinoma (experimental group) and 10 normal tissue by the side of colon carcinoma(control group) with immunohistochemical technique (SP), explore the correlation between the expressions of VEGF,COX-2,MMP-9 and clinical and pathological data(such as tumorous size, position and cell differentiation ). Study the correlation in combined expressions of three proteins, combined expressions of two proteins, simple expression of single protein to the depth of invasion and lymphatic metastasis in colon carcinoma. Logistic monofactor analyze whether the expressions of VEGF,COX-2,MMP-9 proteins are influential factor to the depth of tumorous invasion and lymphatic metastasis in colon carcinoma. Logistic multifactor analyze what are independent risk factors among the expressions of VEGF,COX-2,MMP-9 to the depth of tumorous invasion and/or the lymphatic metastasis in colon carcinoma.Result:1. The expressions of VEGF,COX-2,MMP-9 in 50 colon carcinoma tissue: the positive rate of VEGF,COX-2,MMP-9 are respectively 74%(37/50),72% (36/50),66.%(33/50); In 10 normal tissues by the side of colon carcinoma are respectively 10% (1/10),0% (0/10),10% (1/10). There is significant difference between two groups.2. There is a correlation in the expressions of VEGF,COX-2,MMP-9 proteins in colon carcinoma to depth of invasion and lymphatic metastasis. (P<0.05 ); But there is no difference to age, sex, location of tumor, tumorous diameter, cell differentiation and CEA.3. The spearman analysis indicates that there is a correlation among VEGF,COX-2,MMP-9 in colon carcinoma; There is positive correlation between the combined expressions of VEGF,COX-2,MMP-9 and the depth of invasion and lymphatic metastasis. Furthermore, there is more correlative in combined expressions of three proteins than that of two proteins than that of single protein.4. Logistic monofactor analysis indicates that the expressions of VEGF,COX-2,MMP-9 proteins are correlated to the depth of tumorous invasion and lymphatic metastasis in colon carcinoma. Logistic multifactor analysis indicates that both the expressions of VEGF and MMP-9 proteins are independent risk factors to the depth of tumorous invasion( OR value respectively 5.578 and 9.691), and both the expressions of COX-2 and MMP-9 proteins are independent risk factors to the lymphatic metastasis in colon carcinoma.Conclusion: Colon carcinoma with positive expression of VEGF,COX-2,MMP-9 proteins are easier to promote invading and happen lymphatic metastasis; VEGF,COX-2,MMP-9 proteins play an important role in the mechanism of colon carcinoma invasion and metastasis. They can be used as parameters for the invasion and metastasis of colon carcinoma in molecular biological level.
