The Experimental Study Of Taitaigubao Granules In Treating Postmenopausal Osteoporosis

Objects:This study is aimed at evaluating the therapeutic effect of Taitaigubao Granules and further exploring its action mechanism in treating postmenopausal osteoporosis,by its pharmacodynamics studies.Methods:Four types of animal models applied for studying the major pharmacodynamics were as follows:three rat models of osteoporosis,including ovariectomized,retinoic acid-induced,naturally postmenopausal rat models,and a hydrocortisone induced kidney-deficiency mice model.After 2 months of continous administration of Taitaigubao Granules in the ovariectomized rat model of osteoporosis operated 3 months ago and 40 days of continous administration of Taitaigubao Granules in the retinoic acid-induced rat model of osteoporosis injected 15 days ago,the systematically and vertebrally skeletal density and ALP,TRACP,BGP,E₂,CT and PTH-M concentration in blood serum obtained from the arteria cruralis in each group were measured and compared.After 1 months of continous administration of Taitaigubao Granules in the naturally postmenopausal rat model of osteoporosis,E₂,FSH,LH,ALP and TRACP in blood serum obtained from the arteria cruralis were measured,the wet weight and organ indexes of uterus, adrenal gland and ovaries were recorded.After 14 days of continous administration of Taitaigubao Granules in the hydrocortisone induced kidney-deficiency mice model, the body temperature,spontaneous activities,swimming time at low temperature,the wet weight and organ indexes of thymus,ovaries,uterus,spleen and liver were compared.Statistical analysis was carried out by using the SPSS statistical package (version 11.5).Results:1.Taitaigubao Granules caused a significant increase in the systemic and spinal bone mineral density of the ovariectomized rats and the retinoic acid-induced rats,improved the maximal load of femoral bones and lumbar spines,adjusted levels of blood ALP,BGP,CT and PTH-M,and evalated blood E₂.2.Taitaigubao Granules could obviously alleviate the symptoms of naturally postmenopausal rat model of osteoporosis,increased their uterus organ indexes and levels of blood E₂ and ALP, decreased the blood STRACP,and showed a trend of decreasing the levels of blood FSH and LH and improving aging indexes.3.Taitaigubao Granules evidently relieved the symptoms of kidney-deficiency induced by hydrocortisone,elevated the temperature of kidney-deficiency mice markedly,boosted their spontaneous activities and increased their organ indexes of ovary and uterus.Conclusion:Taitaigubao Granules is in possession of tonifying kidney to strengthen bone,anti-aging,which suggests it is effective in treating women's postmenopausal osteoporosis.It is estimated that the mechanism may be the estrogen-like effect and adjustment of the hormone levels related to levels calcium and phosphorus metabolism as a whole for balance between bone formation and absorption, recoupling process of bone remodeling and achiving the propurse of treating postmenopausal osteoporosis.
