Objective:Investigate the different expression of specific protein 1 (Sp1) between normal renal tissue and renal carcinoma, and its role of growth and apoptosis in 786-0 cell.Methods:1. Investigating the expression of specific protein 1 (Sp1) between normal renal tissue and renal carcinoma by the way of Weston Blot.2. We make use of Weston Blot and RT-PCR to check the DNA and mRNA of Sp1 in 786-0 cell which is delt with Mithramycin A.3. We picture the growth inhibition of 786-0 cell by the way of MTT method while the Sp1 is down regulated.4. We picture the apoptosis of 786-0 cell by the way of Hoechst coloration while the Sp1 is down regulated.Result:1. Sp1 protein is high expressed in 8 cases of renal carcinoma, whileit is not or less expressed in normal renal tissue.2. The protein of Sp1 is less expressed in 786-0 cell which is dealtwith Mithramycin A after 24 hour than that in comparison. 48 hour later, both the mRNA and protein are not presented.3. The growth of 786-0 cell is inhibited through the 24h effect ofMithramycin A. The inhibition is obvious in 24h.4. The apoptosis of 786-0 cell which dealt with Mithramycin A appearsin 24h. The apoptosis is notable in 48h. Conclusion:Wo research present that the protein of Spl is highly expressed in renal cell carcinoma. In 786-0 cell, the growth inhibition is obvious and apoptosis appears through Sp1 which is affected by Mithramycin A.
