Objective:To study the expression and significance of the mitochondrial protein,Bcl-2,Bax,COX-2 and correlations each other in the renal cell carcinoma.To find the mechanism in the tumorigenesis and development.Methods:The expression of the mitochondrial protein,Bcl-2,Bax,COX-2 were detected by immunohistochemistry in 9 cases of the renal oncocytoma and 6 cases of Chromophobe carcinoma and 23 cases of the Clear cell carcinoma as experiment group,respectively and 12 cases of the normal renal tissue as control.Results:1.The expression of the mitochondrial protein:The expression of MAB1273 had statistic difference compaired experiment group with the normal renal tissue(P=0.009),There was difference among the experiment group(P=0.022),It was much higher in the renal oncocytoma than the Clear cell carcinoma(P=0.007),There was no differrance betweeen the Chromophobe carcinoma and the Clear cell carcinoma(P=0.280).There was no differrance betweeen the renal oncocytoma and the Chromophobe carcinoma(P=0.164).The expression of the mitochondrial protein (MAB1273) did not show significant correlations with the clinic pathological parameters.2.The exprssion of the Bcl-2,Bax:Our study found that the expression of the Bcl-2 was higher in all the renal carcinoma groups than in the normal renal tissue.The difference was significant(P=0.008).Its expression had no significant correlation with the subtype(r=-0.175,P=0.294).In the early stage of the RCC,the positivity(Ⅰ+Ⅱ)(18/19)of the Bcl-2 was significant higher than of that in the advanced stage(Ⅲ+Ⅳ)(4/10)(x~2=15.570,P=0.000).The expression of the Bcl-2 had negative correlation with the stage(r=-0.642,P=0.000),it was higher in the low grade RCC(Ⅰ+Ⅱ)(19/21) than in the high grade RCC(Ⅲ+Ⅳ)(3/8)(x~2=9.900,P=0.002),the expression of the Bcl-2 had negative correlation with the grade(r=-0.510,P=0.001).The expression of the Bax was no difference in all the groups(P=0.240).The expression of the Bax was no correlation with the stage and grade(P>0.05).We found in the experiment,in the early stage of the RCC(Ⅰ+Ⅱ),the ratio of the Bcl-2/Bax>=1(17/19)was significant higher than that(4/10)of in the advanced RCC(Ⅲ+Ⅳ)(x~2=10.205,P=0.001).The ratio of the Bcl-2/Bax>=1 was negative correlation with the stage(r=-0.518,P=0.001).In the low grade RCC(Ⅰ+Ⅱ)the ratio of the Bcl-2/Bax>=1(17/21)was higher than that of in the high grade RCC(Ⅲ+Ⅳ)(3/8)(x~2=6.363,P=0.012).Bcl-2/Bax>=1 was negative correlation with the grade(r=-0.409,P=0.011).The rank correlation displayed,the expression of the mitochondrial protein was positive correlation with the expression of the Bcl-2(r=0.350,P=0.013),there was no significant correlation between the expression of the mitochondrial protein and Bax(P>0.05).The expression of the Bcl-2 was negative correlation with the Bax (r=-0.293,P=0.039).3.The expression of the COX-2:The expression of the COX-2 was higher in the renal cell carcinoma groups than in the normal renal tissue(P=0.012).Among the renal cell carcinoma groups the expression of the COX-2 was also different(x~2=7.610,P=0.022),it was much higher in the renal oncocytoma than in the renal clear cell carcinoma(x~2=7.240,P=0.007),the rank correlation displayed the expression of the COX-2 and the Bcl-2 was positive correlation(r=0.279,P=0.049).The expression of the COX-2 was higher in the low grade RCC(18/21) than in the high grade RCC(3/8)(x~2=7.940,P=0.005).And it was negative correlation with the grade(r=-0.457,P=0.004).Conclutions:1.In the RCC,the expression of the mitochondrial protein was significant higher than the normal renal tissue,it indicated the mitochondrial protein might be related with the tumorigenesis of the RCC.It could be the increase of the oxidative stress to lead to the reduced mitochondrial oxidative phosphorylation,mitochondrial proliferation,so that compensated for the reduced the oxidative phosphorylation.Otherwise our experiment displayed the mitochondrial protein was positive correlation with the Bcl-2,it indicated the overexpression of the Bcl-2 might contribute to the release of the mitochondrial protein.So mitochondrial protein could take part in the cell apoptosis in the RCC.2.Our exprement displayed the expression of the Bcl-2 was higher in the RCC than in the normal renal tissue,and mitochondrial protein was positive correlation with the Bcl-2,but there was no significant difference the expression of the Bax,Bcl-2 and Bax was negative correlation,it suggested the expression of the Bcl-2,Bax might take part in the tumorigenesis of the RCC together,suppressed and promoted respectively.We found in the experiment,in the early stage of the RCC(Ⅰ+Ⅱ),the ratio of the Bcl-2/Bax>=1 was significant higher than that of in the advanced RCC(Ⅲ+Ⅳ). The ratio of the Bcl-2/Bax>=1 was negative correlation with the stage.In the low grade RCC(Ⅰ+Ⅱ) the ratio of the Bcl-2/Bax>=1 was higher than that of in the high grade RCC(Ⅲ+Ⅳ),the ratio of the Bcl-2/Bax>=1 was negative correlation with the grade.It suggested the Bcl-2,Bax affected the biological behaviour together.These results indicated the mitochondrial protein,Bcl-2,Bax took part in the tumorigenesis,development.the Bcl-2/Bax could be helpful to evaluate the the grade and prognosis of the RCC.Of course it was need to follow up to confirmed the significance and the value.3.The expression of the COX-2 was higher in the renal cell carcinoma groups than in the normal renal tissue,the expression of the COX-2 and the Bcl-2 was positive correlation.It suggested the COX-2 promoted the RCC tumorigenesis,it could be by up-regulation of the Bcl-2,so inhibited the apoptosis of the tumor cell.The study idicated COX-2 and the mitochondrial protein increased in the RCC,suspected the overexpression of the COX-2 was by the up-regulate of the Bcl-2,so promoted the release of the mitochondrial protein to inhibited the apoptosis of the tumor cells.
