Objectives:Platelet-derived growth factor that is secreted by a variety of cells is a kind of important mitosis agent and chemotactic agent of the connective tissue. PDGF-B and the development of tumors have a close relationship. As he gold standard of tumor angiogenesis assessment, microvascular density can be used to count the tiny blood vessels in the part of most densely populated parts of tumor blood vessels.It is closed related with the malignant behavior?relapse and metastasis of tumor. The present study was to detect the correlations among platelet-derived growth factor expression,microvessel density and multiplepathological parameters in kidny cancer and analyze the significance of PDGF-B expression and MVD in patients with kidny cancer.Material and Methods:Data were collected from 137 operated kidney cancer patients from 1988 through 2006. The expression of PDGF-B and CD34 in different subtype of renal cancer specimens was detected by immunohistochemistry. The correlations among PDGF-B expression?MVD and multiplepathological parameters were then analyzed by ?2-test?Fisher' s exact probability?t-test. Statistical significance was set asp <0.05.Results:1. The expression of PDGF-B was different in different subtype of renal cancer :63.5% in clear cell renal cell carcinoma(CCRCC) tissues; 42.2% in papillary renal cell carcinoma(PRCC) tissues and 48.3/% in Chromophobe renal cell carcinoma(ChRCC) tissues.2. The expression of MVD was different in different subtype of renal cancer : 61.79±34.83 in clear cell renal cell carcinoma tissues; 21.18 ± 13.56 in papillary renal cell carcinoma tissues;20.45 ± 12.62 in Chromophobe renal cell carcinoma tissues.3. The expression of PDGF-B was associated with Tumor size, T-stage in CCRCC(P<0.05) . but it was not associated with with Sex,Age , Furhman classification,metastasis. but with Furhman grade level increases, the expression of PDGF-B increased obviously.4. The expression of MVD was associated with Tumor size ,T-stage and metastasis in CCRCC (P<0.05) . Although it was not statistically significant between Furhman classification and the MVD value,but with Furhman grade level increases, the value of MVD decreased obviously.5. The expression of PDGF-B and MVD was not associated with Sex,Age,Tumor size,T stage,Furhman classification,metastasis in PRCC and ChRCC (P>0.05).6. The MVD values were correlated with the the expression of PDGF-B in CCRCC,but they were not correlated with the the expression of PDGF-B in PRCC and ChRCC.Conclusions:1. The expression of PDGF-B and MVD was different in different subtype of renal cell carcinomas.2. PDGF-B can promote tumor angiogenesis, accelerat tumor cell growth, promot tumor metastasis.3. PDGF-B may become the new clinical prognostic indicators of clear cell renal carcinoma.4. The treatment effect of PDGFR inhibitors on clear cell carcinoma may be superior to the chromophobe renal cell carcinoma and papillary cell carcinoma.
