The Protective Roles Of Astragalus On Acute Liver Injury Induced By CCl₄ In Dogs

With the increase in the number of dogs, liver injury is gradually becoming a common veterinary clinic and frequently-occurring disease in dogs. Liver cell injury was the most typical and common disease in liver injury, which will be chronic liver disease without treatment .Therefore, prevent and treat liver injury timely and effective is an important part of the liver injury. However, the veterinary clinical research on liver disease in dogs is still rare now , there is still not very good way in the treatment of liver injury. Therefore , it has an important clinical practical significance to the dogs acute liver injury pathological process,the changes in the reaction process research and the search for a better method of treatment.In the veterinary clinical practice and animal experimental studies demonstrating the many herbs on liver cell injury has obvious protective effect, the treatment of liver disease has become an important direction. But in the veterinary field, report about Chinese medicine treatment of canine liver injury was also rarely, and most experiment was just confirmatory test ,which could not explain the mechanism of prevention or treatment of liver injury.Astragalus is the dry root of legumes such as Mongolia Astragalus or Astragalus membranaceus .It's sweet and moderate. Astragalus can promote immunization, improve the activity of macrophages, regulate the blood sugar, delay aging of cells . Astragalus can improve the anti-injury and infection capacity. This experiment was made on the basis of the prevention and treatment experience of liver injury in veterinary clinic.Dogs was been used in this experiment to be observed on the role of Astragalus for the antioxidant indicators,the protective effect for liver function,the liver tissue paraffin sections in the acute liver injury modle induced by CCl₄. Which was made to provide a theoretical reference for the treatment of liver damage in dogs using Chinese medicine. Therefore, the study can provide better theoretical for the treatment of liver injury,reduce the economic losses,enhance social and economic benefits.Healthy dogs weighed 4-6kg were used as experimental animals, and were randomLy divided into the control group and the experimental group , there were 5 dogs in each group. the acute hepatic injury model was prepared by injection of 50% CCl₄ plant oil solution with the dose of 1mL/kg BW, the experimental group was infused. Astragalus extracts(1g crude drug/mL drug liquid) with dose of 1mL/kg BW, the control group didn't receive any treatment. Blood was picked from jugular vein and serum was separated, then the content of MDA,T-AOC and the enzyme active of GSH-PX,AKP,ALT,AST in serum was measured. On the seventh day after the modle was made successfully, the dogs were anesthesiaed with"SUMINXIN". Aseptic ripped abdominal cavity to the left of the liver roughly the same location, fixed with 10% neutral formalin to made the Paraffin sections. The results showed that:1. During the experiment, the dogs of the experimental group infused with Astragalus extracts were energetic and have good appetite without no obvious side effect , while the dogs in control group showed listless, diarrhea with blood in stool. Those shew that Astragalus extracts can improve the clinical signs of dogs with acute hepatic injury model.2. Compared with the control group, the enzyme active of AKP and the content of MDA in the experimental group arose less than the control group's, the enzyme active of GSH-PX and the concentration of T-AOC in the experimental group elevated less than the control group's. These indicated that Astragals extracts could enhance the level of anti-oxidation,induced the injury made by carbon-tetrachloride.3. Compared with the control group, the enzyme active of ALT,AST arose less than the control group's, the liver cell degeneration and necrosis induced by liver injury have been improved significantly .It showed that the Astragals extracts could improve the reduction of liver function caused by CCl₄ , improve the pathological changes such as liver cell degeneration and necrosis, protect and restore the damaged liver cells .
