Objective:1)To study the influence of spray drying on the anti-oxidation of grape seed extract.2)To study the protective effects of grape seed extracts against the immunological and chemical liver injury in mice.Methods:1)Screening the grape seed from the drags of a winery in xinjiang,After defating completely by DAB-6,refluxing extract by EtOH,decompressed concentration,extracta siccas powder was made by two kind of drying methods.Compare the anti-oxidation of the two extracta siccas powder including reductive ability,and scavenging activity on DPPH and hydroxyl radical.2)60 male mice were randomized into control group,model group group,low dose group,middle dose group and high dose group bifendate group;The control group were intravenously injected the isotonic Na chloride 0.2ml,other groups were intravenous injected Bacillus Calmette-Guerin 0.2ml(contain5×10~7viable organism)isotonic Na chloride suspension. Three experimental groups received intragastric administration with grape seed extracts of 50,250,500mg/kg respectively,which lasted 12 days.After one hour of last intravenous injection,the control group was intravenous injected isotonic Na chloride 0.2ml other groups were intravenous injected lipopolysaccharide(LPS)7.5μg/ each mice.12 hours of the last intravenous injection.72 male mice were randomized into control group,model group,low dose group,middle dose group and high dose group bifendate group;Every morning the control group were intragastric administration vegetable oil(0.2ml/10g),other groups were intragastric administration 2%carbon tetrachloride oil solution(0.2ml/10g)Every afternoon Three experimental group were intragastric administration grape seed extracts with 50,250,500mg/kg respectively,the control group and model group were intragastric administration isotonic Na chloride last 10days 10hours of last intragastric 3)alanine a minotransferase(ALT)and aspartate amiontransferase(AST)activities in serum;superoxide dismutase(SOD)activities, malondialdehyde(MDA)content in liver homogenate were assayed and hepatic pathological changes were observed.Results:1)It showed that difference of bioactivity between these two kinds of extracta siccas powder makes no statistical meaning.2)Grape seed extracts(50,250,500mg/kg)decreased the coefficient of liver and spleen and the content of MDA(P<0.05);grape seed extracts(50,250,500mg/kg)increased the activity of liver homogenate SOD(P<0.05);grape seed extracts(250,500mg/kg)decreased the activity of serum ALT and AST(P<0.05).grape seed extracts improved liver histopa-thology injuries.Grape seed extracts procyanidin decreased the content of MDA (P<0.01);grape seed extracts procyanidin increased the activity of liver homogenate SOD(P<0.01);grape seed extracts decreased the activity of serum ALT and AST(P<0.01).Grape seed extracts improved liver histopathology injuries.Conclusions:1)These results suggested that drying spray have no influence on the anti-oxidation of grape seed extracts,so it can be used in industrial production.2)Grape seed extracts showed protective effects on the immunological liver injury in mice.3)Grape seed extracts showed protective effects on the acute chemical liver injury induce by carbon tetrachloride in mice.
