BackgroundResistin is a newly identified adipocytokine that has been proposed to be a link between obesity and type 2 diabetes based on animal studies。In human resistin is secreted mainly by leukocytes,especially by monocytes.Now,its physiological and clinical role has not been well-defined.Recent data have shown that resistin plays an important role in insulin resistance,obesity,type 2 diabetes and atherosclerosis,which are risk factors of cerebral infarction.To investigate whether the -420C>G polymorphism of resistin gene is associated with cerebral infarction and its clinical characteristics,we conducted a case-control study.Objective1.To investigate the relationship between the SNP -420C>G of resistin gene and cerebral infarction.2.To study whether the SNP -420C>G of resistin gene is associated with the risk factors and clinical characteristics of cerebral infarction.MethodsThe single nucleotide polymorphism(SNP)of resistin was demonstrated by means of PCR and digested by Restriction Endonucleases in the 258 cases of cerebral infarction and 231 healthy controls.The study group was divided into two subgroups, cerebral thrombosis group(176cases)and cerebral embolism group(82cases).The measurement data was analyzed statistically by t-test and enumeration data by chi-square test.The disease risk factors were analyzed by binary logistic regression.Results 1.There were three genotypes at the site of-420 of resistin gene:CC,CG and GG..2.There were significant differences of CC,CG and GG genotypes(P=0.014)and C/G allelic frequencies(P=0.002)between patients with cerebral thrombosis and healthy subjects,while there were no significant differences of CC,CG and GG genotypes(P=0.954)and C/G allelic frequencies(P=0.986)between patients with cerebral embolism and healthy subjects.3.Subjects with cerebral infarction show a significant association between cardiovascular disease and SNP-420C>G(P=0.014).The CG+GG genotype had a higher frequency.4.The association between SNP-420C>G and type 2 diabetes or insulin resistance showed no significance.5.Binary logistic regression analysis to cerebral thrombosis group showed OR value was 0.5.It meant that the risk of cerebral infarction doubled the CG+GG group than that of CC group.Conclusion1.The SNP -420C>G of resistin gene is implicated in the pathogenesis of cerebral thrombosis.The frequency of G alleles in -420 C>G genotype is higher obviously in the group of cerebral thrombosis.2.There are no significant association between SNP-420C>G and cardiovascular disease.3.There are no significant association between SNP-420C>G and type 2 diabetes or insulin resistance.4.The SNP-420C>G of resistin gene may play a role in cerebral infarction and cardiovascular disease probably through accelerating atherosclerosis.5.The results show the Systolic blood pressure of CC type is higher than that of (CG+GG)type.It need to be further confirmed by larger samples.6.The results suggest that the -420C>G polymorphism of resistin gene may be a risk factor for cerebral thrombosis.Significance The detecting and studying to SNPs of resistin gene,may predict the stroke risk factors early.Timely prevention and intervention can lead to early genetic diagnosis and gene therapy of stroke.
