| Objective:To observe the effect of Nourishing Yin Antitoxic(YC) on the immunohistochemistry of pituitary ACTH cells in Yin deficiency rats,and To observe CORT,SOD and MDA of kidnuey Yin deficieney rat to prove the curative effect of YC.so it was to provide the basis for clinical application of YC.Methods:Experiment 1:Effect of Nourishing Yin Antitoxic Capsule on the Immunohistochemistry Of Pituitary ACTH cells in Yin Deficiency RatsFourty SD rats were randomly divided into four groups:normal control group,Yin deficiency model control(YD)group,YC group,Kangdu group (Daxiaoji,Huaihua).After pretreatmented by chinese medicine and administratior 7 days,the SD rats were killed.YC Capsule power is repulverized,then filted 100-meshwork,add distilled water,and make it for 5 mg/mL suspl with sonic oscillation.Kangdu group is prepared for suspl of 5 mg/mL with distilled water Before used.The dosage of YC and Kangdu group used in the rats is 25 times more than adults,7.5g/d.The average weight of each rat is 200g,so the dosage used in every rat is 0.5g.The dosage is diluted to 0.2g/ml with distilled water and administration of each rat is p erformed every moring with concentration of 10ml/kg for 3 days.The control group and YD group are lavagd with distilled water,the dosage is 2ml/d for 3days.From the forth day,all grouos except control group are lavagd with hydrocortisone every 17:00 for 4days,the dosage is 50 mg/kg,and the control is lavaged with distilled water,2 ml/d.the rats are sacrificed at the same period after 9-day pre-disposal treatment of Chinese crude drug,and then ext the pituitary gland in 2mins,fix it with 4%paraformaldehyde, paraffin imbedding,stain ACTH with method of immunohistochemistry.Experiment 2:The Effect of CORT,SOD and MDA Level in Blood by Nourishing Yin Antitoxic Capsule and its separate components for Experimental Kidney Yin Deficiency in RatsFourty SD rats were randomly divided into four groups:normal control group,Yin deficiency model control(YD)group,YC group,Kangdu group(Daxiaoji,Huaihua).After pretreatmented by chinese medicine and administration 9 days,the SD rats were killed.To observe CORT,SOD and MDA of kidnuey Yin deficieney rat to prove the curative effect of YC.The process of duplicated model is as experiment one.All groups are anaesthesiaed using 846(1ml/kg ) at 8:00 to 09:00 after 1-day complete of duplicated model(the ninth day),then get blood from the cordis,and determine the contents of SOD,MDA,CORT using xanthinoxidase colorimetric method,thio-barbital sodium colorimetric method,radio-immunity(RIA) respectively.Results:1,Compared with the normal group,the contens of ACTH were higher in YD group,which was(157±67.8)(P＜0.05),Compared with the YD group,the contens of ACTH were lower in YC group,which was 102±37.5(P＜0. 05).however,there is not significantly different in separate components of YC,which was 145±47.8.2,The contens of CORT and MDA were higher than YD group(9.10±1.70μg/L,P＜0.05),Meanwhile the SOD were lower than YD group (520.75±126.87μmol/L,P＜0.05),the contens of CORT(6.01±2.11)μg/L and MDA(10±1.40)μmol/L were lower than YD group,Meanwhile the SOD were lower than YD group(801.10±41.57μmol/L,P＜0.05).however,there is not significantly different in separate components of YC,indicating the reasonability of compatibility of medicine in science of formulae.Conclusion:In this experiment,the cell expression of ACTH in Yin deficiency rats was lower.The YC can decrease the expression of pituitaryACTH,andimprovehy-pothalamic-pituitary-adrenal(HPA)axis, and diminish the side effect of glucocorticosteroid.The medicine has very obviously protective action to Kidney Yin deficiency by the way to influence the content of SOD and MDA in the blood. |
