An Experimental Study On The Effects Of Yougui Pill On T, LH And CORT In Rats With Kidney-yang Deficiency Based On The Theory Of "Yin Yang"

Objective:To explore the effect of Yougui Pill and its disassembled prescriptions on T,LH and CORT in serum of Kidney—Yang Rats Deficiency.Methods:A total of 50 male SD rats were selected and randomly divided into normal control group,model group,nourish yin group,tonifying yang group and the whole recipe group,with each group containing only 10 rats.Except the normal group,the other four groups were treated with intramuscular injection of 2.5 mg/100 g at hind hydrocortisone 14 d to make the model of kidney yang deficiency,according to physiological status of kidney yang deficiency and urinary 17-OHCS expression decreased in judging model success.Model second days after each group were given corresponding drugs by gavage for 14d(dosage according to body surface area method conversion was 5 times of adult and dosage by intragastric dose of 1 ml / 100g),normal group and model group were given normal saline by intragastric administration of 14 d.After discontinuation of 2d were obtained by heart blood serum and removal of testes made.The body weight and testis weight of rats were recorded,and the testis index was calculated.The contents of T,LH and CORT were determined by ELISA method.Observed the testicular fine structure change under light microscopy.Results:1.The rats weight and index of testis The three treated groups were able to increase the body weight and testis index of rats with kidney yang deficiency syndrome,with statistical significance.(P < 0.05)The whole recipe group the most obvious effect,while nourish yin group increased relative weight of the rats were stronger(P < 0.05),tonifying yang group to enhance strong testis index(P < 0.05).2.Determination of T,LH and CORT by ELISA Three groups treatment group to improve kidney Yang deficiency syndrome in rats serum T expression level of the whole recipe group effect is best,tonifying yang group set times,between groups was statistically significant difference(P < 0.05);Three groups treatment group to improve the expression of kidney Yang deficiency syndrome rats serum LH level,nourish yin group effect is best,the whole recipe group set times,difference between groups was statistically significant(P < 0.05);Three groups treatment group to improve kidney Yang deficiency syndrome in rats serum CORT level,the whole recipe group have certain regulation effect(P < 0.05),nourish yin group and tonifying yang group has no obvious effect(P > 0.05),there was no statistically significant difference between groups(P > 0.05).3.Testicular fine structure under light microscope Relative to the normal control group,nourish yin group all levels of spermatogenic cells reduced,spermatogonia,primary spermatocytes have pyknosis signs,including the quality of sperm cells have disappeared,sperm count decreased obviously,and the form of chaos.Relative to the model control group,has certain effect;tonifying yang Group and the model control group had little difference,the number of spermatogenic cells were arranged orderly.Compared with the model control group,the morphology and structure of the control group were normal,and the number of sperm cells and sperm increased.Conclusion:The whole Party Yougui Pill on kidney yang deficiency rats body weight,testis index,serum T,LH and CORT expression and testicular micro structure produce the positive influence in the party,nourish yin in the promotion of kidney yang deficiency rats body weight and increase the concentration of plasma LH in the regulation effect is better,tonifying yang increase the plasma concentration of T and improve the testicular index,repair the damaged testicular tissue in the regulation effect is better,nourish yin and tonifying yang had no obvious effect on increasing the expression of CORT in plasma.
