Objective:To observe the effect of Nourishing Yin and Antitoxic CapsuleⅡ(YCⅡ) ,a preparation of traditional Chinese medicine, on oxidative injury caused by X-ray radiation at a dose of 6 Gy ,which makes SD rats haematogenesis injury acutely,so it was to provide the basis for clinical application of YCⅡ.Methods:Experiment 1: the effect of YCⅡon oxidative injury by radiothe- rapy.Thirty clean adult male SD rats(200±20g) were randomly divided into three groups with ten rats each: the normal group (A), the radiation group (B), the treatment group (C). The rats in group B and C were exposed to whole body by X-ray radiation at a dose of 6 Gy. All the rats were treated by intragastric administration respectively at a dose of 2.5ml: group A and B with Sodium Chloride; group c with solution of YCⅡ(0.2g/ml) . At the eighth day after radiation the rats were executed and the blood samples (3-6ml) were taken from the hearts of the rats. The level of serum malondialdehyde (MDA),activity of serum superoxide dismutase (SOD),activity of serum catalase(CAT)and the total ant oxidative capacity (T-AOC) were measured. WBC quantity was counted to evaluate whether the rat model succeeded. The pathological changes of bone marrow of rats were observed by light microscopy. And the spleen and thymus index were counted.Experiment 2 : the effect of YCⅡon adrenal cells ultramicrost- ucture injured by radiotherapy.Thirty adult male SD rats were randomly divided into three groups with ten rats each: the normal group (A), the radiation group (B), the treatment group (C). The rats in group B and C were exposed to whole body by X-ray radiation at a dose of 6 Gy. All the rats were treated by intragastric administration respectively at a dose of 2.5ml: group A and B with Sodium Chloride; group c with solution of YCⅡ(0.2g/ml). At the eighth day after radiation the rats were executed and the adrenal tissue sample was dislodged and been made tissue slice. Pathological changes of adrenal cell of rats were observed by electron microscope.Results:1,YCⅡdelayed significantly the decline of the quantity of RBC and WBC in circulation due to ionizing irradiation (P < 0. 01), impro- ved the index of thymus and spleen in ionizing irradiation rats(P < 0. 01) ,increased the activity of super- oxide dismutase(SOD),catalase (CAT)and the total antioxidative capacity (T-AOC), decreased the level of malodialdehyde (MDA) in the rats'serum . By light microscopic a lots of adipocytes were observed; hematopoietic cells swelled and degenerated; cell nucleus were thick and pyknosised; cell junction was not clear in group B .And in group C a fewer adipocytes and deg- enerating cells were observed.2,electron microscope show that in radiation group cel- lular swelling was obvious in many cells; lipid droplet def- luxioed; some chondrosomes concentrated and some chondrosomes swelled; endocytoplasmic reticulum dilate; many myelin figure emerged; cell nucleus heterochromatin increased markedly arr- anging at the edge of nuclear membrane. And in group C cell nucleus heterochromatin increased comparing with in group A; chondrosomes swelled lightly and chondrosomes vacuolization was not marked; few lipid droplet was founded ,which shows that the cytoactive of adrenal was at a lower level comparing with in group A. Conclusions:YCⅡcan improve anti-oxidative capacity; decrease oxidative injuries induced by X-ray irradiation in rats; and at the same time preserve or shield adrenal cells from radiation injury at the stress of exposing to X-ray, so exhaustion of organism can be avoided.
