Relationship Between SNP Of Apolipoprotein Genes And Efficacy Of Traditional Chinese Medicine

Coronary heart disease (CHD) is significantly harmful for the patient, Qi deficiency blood stasis (QDBS) is a common type of CHD, and can be treated by Qi Shen Yi Qi drop pill. Apolipoprotein (APO) genes are closed related to CHD, but the relationship between SNPs of APO genes and Qi deficiency blood stasis and efficacy of traditional Chinese medicine (TCM) haven't been reported.There were 49 patients wi^th QDBS taken Qi Shen Yi Qi drop pill for one month. The blood samples were taken before and after the treatment, and then DNA was extracted from the white blood cells. The first extron of APOA1 gene, the third extron of APOA4 gene and the four^th extron of APOE gene were amplified by polymerase chain reaction (PCR). The PCR conditions were as follows: denaturation for 3 minutes at 95℃, and 35 cycles of denaturation for 1 minute at 94℃, annealing for 50 seconds at 65℃for APOA1 gene, for 50 seconds at 55℃for APOA4 gene and APOE gene, and final extension for 10 minutes at 72℃. The product lengths are 767 bp, 1148 bp and 478 bp, repectively. After being sequenced the SNPs were detected by comparing wi^th database. The density of SNPs of APOA1, APOA4 and APOE are 13.33%, 0.08% and 0.93%, repectively.Seven significative SNPs are found, of which, three SNPs relating to only CHD, one SNP only relating to efficacy, two SNPs relating to CHD and efficacy, and two SNPs unrelating to CHD or efficacy. The 276^th mutant in APOA1 gene enhances self-perspiration and BG content. The 295^th mutant in APOA1 gene also increase BG content. The 339^th mutant in APOA1 gene enhances self-perspiration, but decreases contents of TC and VLDL. The 917^th mutant in APOA4 gene also enhances self-perspiration and increases BG content, however, LDL content was decreased. The 2888^th mutant in APOE gene decreases MDA content and enhances the efficacy alleviating mental weariness. The 2903^th mutant in APOE gene significantly enhances the efficacy alleviating self-perspiration and chest pain, but increases content of BG and apoB.APO genes affect CHD and efficacy of TCM, this research tries to reveal the relationship between SNP and efficacy of TCM. It is significant to developing TCM and instructing rational drug administration.
