Background and ObjectivesCHD(Coronary Heart Disease)falls within the scope of multi-genetic disease. Therefore,in compliance with the rapid development of human genome plan,the orientation and identification of CHD-related genes have been hot issues in medical research.In pathology,it is indicated that the fracture of atheromatous plaque in coronary artery,platelet aggregation and thrombosis,all result in unstable angina pectoris and myocardial infarction.Recently,evidences have showed that due to inflammation involving,the begetting of artery thrombus disease is in close relation with development of it.Meanwhile,the consistence of inflammation marker either in healthy people or CHD sufferer is to do with CHD in the future.Macrophage migration inhibitory factor(MIF)is secretion factor of macrophage,which plays important role in inflammatory reaction,and is considered as the key medium for arteriosclerosis.Researchers have discovered MIF-173 relates to self-immunity diseases,however,few researches explore whether it has an impact on CHD.The purpose of this study was:to known the distribution of macrophage migration inhibitory factor(MIF)-173 gene single nucleotide polymorphism in Chinese people and the relationship with CHD.Material and Methods1.SubjectCHD(+)group:152 CHD patients(96 men and 56 women,mean aged 62.53±10.01 years) were consecutively collected from the hospitalized in the department of cardiology,Sir Run Run Shaw hospital(the affiliated hospital of medical college,Zhejiang University).All patients were confirmed by coronary angiography.CHD(-)group:175 CHD(-)object(102 men and 73 women,mean aged 62.46±11.69 years)served as a control group.All subjects were consecutively collected from the hospitalized in the department of cardiology,Sir Run Run Shaw hospital.All patients were confirmed have no CHD by coronary angiography.2.Laboratory methods for MIF-173 gene polymorphism1)DNA was extracted from the peripheral blood leukocytes by standard phenol and chloroform method.2)Macrophage migration inhibitory factor-173G/C polymorphisms were detected by polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP).3.Statistic analysisUsing the SPSS11.5 for windows statistical package Count Data were performed by Student' s t-test and measure data were performed by chi-squared test.Genotype and allele frequencies between groups were analyzed by the chi-squared test. Statistical significance was taken as p<0.05.Results1.The authors detected three kinds of genotypes at the MIF-173 locus,and no deviation was observed from Hardy-Weinberg equilibrium.(p>0.05).2.The MIF-173 C allele was detected in 89.1%of normal controls and 75% of patients,while MIF-173 C allele was detected in 0.109 of normal controls and 0.25 of patients.There was significant difference in MIF-173 genotype distribution between patients and controls.3.The genotype distribution of MIF-173 was no difference between single,double and triple vessel disease(x~2=1.34,p>0.05).4.The genotype distribution of MIF-173 was no difference in patients with and without myocardial infarction(MI).Conclusion 1.There is a polymorphism of MIF-173 in Zhejiang Han population.2.C allele may be a susceptibility gene of CHD.3.MIF-173 gene polymorphism may be associated with the degree and type of CHD.4.MIF-173 gene polymorphism may be not associated with gender,hypertension,and diabetes mellitusem(DM).
