Objective: To research the expression of Kai1 and p53 in colorectal carcinomas and study the effect of Kai1 and p53 on the pathogenetic condition of colorectal carcinomas; also study the correlation of Kai1 and p53 in colorectal carcinomas.Method: The paraffin specimens were taken from 56 patients with primary colorectal carcinomas in the Second Affiliated Hospital of Nanchang University from Nov 2006 to Oct 2007. 28 adenomas and 22 normal intestine specimens were used as control. EliVision immunohistochemical method was used to detect the expression of Kai1 and p53 in colorectal carcinomas , adenomas and normal intestine.Results:(1) The expression of Kai1 in colorectal carcinomas,adenomas and normal intestine.Kai1 chiefly located in cell membrane, also in cytoplasm. The protein expression of Kai1 was down-regulation in colorectal carcinomas compared to the adenomas and normal intestine(P<0.01),there was no difference of Kai1 expression between intestine adenomas and normal intestine .The expression of Kai1 was not correlated with the age of patients, the gender of patients, the location of tumour , the tumour size and infiltration degree(P>0.05),but its expression was significantly increased in well and moderately differentiated group, no-lymph node metabasis group, no-distant metabasis group and TNMâ… /â…¡compared to that of poorly differentiated group ,lymph node metabasis group, distant metabasis group and TNMâ…¢/â…£(P<0.05)(2)The expression of p53 in colorectal carcinomas,adenomas and normal intestine.p53 located in cell nucleus. There was obvious difference of p53 positive rate among colorectal carcinomas, adenomas and normal intestine ( P<0.01 ) .The expression of p53 was not correlated with the age of patients ,the gender of patients, the location of tumour ,the tumour size ,infiltration degree and distant metabasis(P>0.05).The expression of p53 was significantly increased in the poorly differentiated group, lymph node metabasis group and TNMâ…¢/â…£. ( P<0.05).(3)The relationship between the expression of Kai1 and p53 in the colorectal carcinomas .When the expression of Kai1 was negative ,there was one negative expression of p53 and 21 positive .When the expression of Kai1 was positive ,there were 12 negative expressions of p53 and 22 positive .There was a negative correlation between the protein expression of Kai1 and that of p53.( rs'=-0.5763,P=0.0000)Conclusions:(1)The abnormal expression of Kai1 and p53 maybe paticipate in carcinogenesis and progression of colorectal carcinomas.(2)There is assistant value in evaluation of pathogenetic condition and prognosis of colorectal carcinomas by combining examination of both Kai1 and p53.(3)The expression of Kai1 may be directly controlled by p53 in colorectal carcinomas.
