Objective: To investigate the correlation KAI1 gene expression with metastatic behavior of human colorectal carcinoma.Method: KAI1 mRNA expression was examined in 45 colorectal primary carcinomas,4 liver metastases, 10 lymph node metastases by self-quantitative reversetransciption-polymenase chain reaction (RT-PCR), 45 normal colonic mucosa tissue samples served as controls. Result: KAI1 expression was higher in primary carcinomas than in normal mucosa, (40/45 ,P<0.01) ,and was inversely correlated with Dukes'stage and lymph node metastasis(P<0.0 1 ).KAI 1 expression was lower in Dukes'C and Dukes' D than in Dukes' B (P<0.05),and more lower in lymph node metastases (P<0.01). KAI1 expressionwas significantly different in primary colonic tumors in the presence and absence of lymph node metastasis.On multivariate analysis,there were statistically significant correlations between KAI1 down-regulation and lymph node metastasis (P<0.05). Conclussions: The up-regulation of KAI1 gene seems to beinvolved in the early stage of colorectal carcinoma .The expression of KAI1 was inversely correlated with lymph node involvement, Dukes' stage .The down-regulation of KAI1 gene are associated with lymph node metastasis of colorectal carcinoma. KAI1 gene acted as a metastasis suppressor gene in human colorectal carcinoma. KAI1 might play some role in predicting the risk for tumor spread , determining treatment and creating new options in prevention of tumor spread or metastasis.
