Because animal models of depression and kidney-yang asthenia couldn't be prepared at the same time.The experiment was designed to rat model of depression and rat model of kidney-yang asthenia.The effect of Complex Prescription Decoction Bushenkangyu on two rat models was observed.Objective:To establish the solitary custody and chronic stress and depression rat model and kidney-yang asthenia rat model,investigate the mechanism and dose-effect relationship of Complex Prescription Decoction Bushenkangyu on two rat models,providing pharmacodynamic basis.Method:PartⅠ:60 healthy male Wistar rat were selscted and divided into 6 groups randomly: group A were blank group;group B were model group;group C were fluoxetine hydrochloride group;group D~F were Complex Prescription Decoction Bushenkangyu low dose group, medium dose group and high dose group.Each group included 10 rats.On the 21st day of Experiment,through the weight of the rat and behavioral testing to prove that the success of modeling.After modeling success,group A,B:distilled water,1ml·100g-1·d-1;group C: Fluoxetine suspension,3.33mg·kg-1·d-1,group D~F:Complex Prescription Decoction Bushenkangyu suspension,0.5g·kg-1·d-1,1g·kg-1·d-1,2g·kg-1·d-1,21d were gavage.Experiment at the end of rat weight,behavioural indicators and monoamine neurotransmitters in the brain content changes.PartⅡ:50 healthy male Wistar rat were selscted and divided into 5 groups randomly:group A were blank group;group B were model group;group C~E were Complex Prescription Decoction Bushenkangyu low dose group,medium dose group and high dose group.Each group included 10 rats.On the 9st day of Experiment,through the weight of the rat and behavioral testing to prove that the success of modeling.After modeling success,group A,B:distilled water,1ml·100g -1·d-1;group C~E:Complex Prescription Decoction Bushenkangyu suspension,0.5g·kg-1·d-1, 1g·kg-1·d-1,2g·kg-1·d-1,9d were gavage.Experiment at the end of rat general condition,weight, weight adrenal gland and the adrenal cholesterol content changes.Result:PartⅠ:1.Rat body weight and behavioural effects:group D,E,F than group B,the weight increase,the increase in sugar consumption,Open-field scoring higher escape latency and distance swimming were significantly reduced,There was statistical significance(P<0.05). 2.Rats brain 5-HT,NE,DA content:group C,F was significantly higher than group B,statistical significance(P<0.05).3.group D,E,F,the different indicators have improved role strong or weak and there were some dose-effect relationship.group F for the optimal group. PartⅡ:1.General state of rats and weight impact:group B,C than group A,weight loss, statistical significance(P<0.01),and the general poor state.2.group D,E than group B,weight gain of more than a statistical significance(P<0.05).3.Rat adrenal gland:group D,E than group B,with statistical significance(P<0.05).4.the adrenal cholesterol content changes:group D,E than group B,with statistical significance(P<0.05).5.group E for the optimal group.Conclusion:1.Complex Prescription Decoction Bushenkangyu can increase weight,improve behavior score,enhance learning and memory,increase 5-HT,NE,DA content for rat model of depression.2.It also can improve general state for rat model of kidney-yang asthenia. Glucocorticoid inhibition caused by rat adrenal atrophy and cholesterol accumulation of glucocorticoid the adrenal injury protection.3.Complex Prescription Decoction Bushenkangyu different doses of anti-depression on rats and rats kidney-yang asthenia affecting certain dose-effect relationship,high dose group and medium dose group role significantly,and them in the difference between not.4.Complex Prescription Decoction Bushenkangyu high dose group and the control group of western medicine fluoxetine hydrochloride considerable effect.
