Objective To investigate interleukin-1A-889 , interleukin-1B-511 (IL-1A-889, IL-1B-511) genotype and its association with the susceptibility of chronic periodontitis and coronary heart disease so as to uncover the underlying etiopathogenesis for improving the diagnostic conclusion and the design of preventive and therapeutic strategies targeting the diseases. Methods every subject was examined on periodontium, then the probing depth (PD) and the clinic attachment loss (CAL) was recorded .DNA was obtained using Chelex-100 from buccal swabs of 44 subjects with moderate to severe chronic periodontitis, 41 subjects with coronary heart disease, 40 subjects with moderate to severe chronic periodontitis and coronary heart disease and 49 matched healthy control individuals, which was from hospital patients in the department of Cardiology and medical outpatients, including subjects with moderate to severe chronic periodontitis and healthy control volunteers, in Department of Stomatology of the first teaching hospital of Xinjiang Medical University. Genotypes of IL-1A-889/NcoI, IL-1B-511/AvaI was analyzed by sequence specific primers-polymerase chain reaction (SSP-PCR) and PCR-restriction fragment length polymorphism (PCR-PFLP) method. Then compared the differences indistribution of genotype. Results (1) There was no statistical differences about indistribution of IL-1A-889 genotype among the four groups (P>0.05). (2) A significant Statistical difference about indistribution of IL-1B-511 genotype was found among the four groups (P<0.05). A more Significant over-representation of IL-1B-511 allele 2 was found in CHD group than healthy controlindividuals, and there was a statistical difference(P<0.0083).(3)A more Significant over-representation of the compound allele 2 of IL-1A-889 and IL-1B-511 was found in CP group than healthy control individuals, and there was a statistical difference (P<0.0083). Conclusion This result suggests that IL-1B-511 allele 2 may be a risk indicator for the susceptibility of CHD, and the compound allele 2 of IL-1A-889 and IL-1B-511 may be associated with chronic periodontitis susceptibility.
