Objective: Adenomyosis is a common gynecologic benign disease,hitherto its pathogenesis has not been elucidated.We investigated what role do 14-3-3 proteins play in the development of human adnomyosis by detecting the expression of 14-3-3 mRNA in normal endometrium,eutopic endometrium and ectopic endometrium with adenomyosis.Method: Tissue samples of eutopic endometrium and ectopic endometrium were obtained by hysterectomy from fifteen reproductive women who were diagnosed as adenomyosis .Tissue samples of normal endometrium were obtained by diagnostic curettage from fifteen reproductive women who were diagnosed as benign tumor of ovary(mature teratoma,cyst) and were in hospital at the same time with those women who were diagnosed as adenomyosis. Tissue samples were stored in liquid nitrogen. All specimens were verified by pathology and separated into three groups: (1) group:normal endometrium; (2)eutopic endometrium;(3)ectopic endometrium. The expression of 14-3-3mRNA was measured by RT-PCR in normal endometrium,eutopic endometrium and ectopic endometrium with adenomyosis. The experimental datas were obtained by ImageTool software. Statistical analysis was performed using SPSS11.5 for windows.Results: The level of every isoform of 14-3-3mRNA in normal endometrium( Control ),eutopic endometrium(EU) and ectopic endometrium(EC) with adenomyosis were as follow:(1)14-3-3θmRNA:14-3-3θmRNA was the highest transcribed in EC group when compared with both of control group and EU group(p<0.01), but there were no significant difference between EU group and control group(p>0.05);(2)14-3-3εmRNA:The transcription of 14-3-3εmRNA in EU group and EC group was dramatically increased when compared with that of control group(p<0.05)but there were no significant difference between EU group and EC group(p>0.05);(3)14-3-3ζmRNA:The expression of 14-3-3ζmRNA in EU group was significantly increased when compared with that of control group and EC group(p<0.01),and EC group had higher expression of 14-3-3ζmRNA than that of control group(p<0.05);(4)14-3-3β,γ,η,σmRNA:With regard to the transcription of of these four isoforms of 14-3-3 proteins,there were no significant difference among Control group,EU group and EC group. (p>0.05).Conclusion:1. Results of our study indicated that 14-3-3mRNA showed a basic expression in normal endometrium,eutopic endometrium and ectopic endometrium with adenomyosis.2.The expression of 14-3-3θ,εandζmRNA in eutopic and(/or) ectopic endometrium with adenomyosis was up-regulation, which suggested those three isoforms of 14-3-3 proteins may be involved in the development of adenomyois.
