BackgroundAdenomyosis refers to benign infiltration and diffused growth of endometrial glands and stroma to myometrial, also known as intrinsic endometriosis. In recent years, studies have found that invasiveness of endometrial cells may be related with the pathogenesis of adenomyosis.Kiss-1 is a tumor metastasis suppressor gene.The study shows that Kiss-1 could inhibit chemical trends and invasiveness of cells, and malignant melanoma, thyroid cancer, endometrial cancer and other tumors are closely related. Heparanase so far is only able to cleavage heparan sulfate proteogly which is the main component of extracellular matrix. Study has confirmed heparanase is related with tumor angiogenesis, cell invasion, adhesion and metastatic potential. Therefore, the study on Kiss-1 and heparanase in patients with adenomyosis is significant for exploring the pathogenesis of adenomyosis. This study explore the pathogenesis of adenomyosis through investigating invasiveness of endometrial cells.Mifepristone is an anti-progesterone drug acting on the receptor level. It plays a role through competition with the receptor and blocking progesterone. Mifepristone can be used for clinical treatment of adenomyosis for improving the symptoms and have a therapeutic effect. This issue that investigated the effects of mifepristone intervention on Kiss-1 and Hpa mRNA expressions in cultured glandular cells from eutopic endometrium in adenomyosis have guiding significance for clinical medicine.Object To investigate the expressions of Kiss-1 and Hpa mRNA in cultured glandular cells from eutopic endometrium in adenomyosis and normal endometrium in control group. To explore the effects of mifepristone intervention on Kiss-1 and Hpa mRNA expressions in cultured glandular cells from eutopic endometrium in adenomyosis. Furthermore , to make out the relationship between Kiss-1 and Hpa mRNA and adenomyosis and the function of mifepristone.Method Culture the glandular cells of eutopic endometrium in adenomyosis and normal endometrium in control group .Then detect the expressions of Kiss-1 and Hpa mRNA in adenomyosis group,mifepristone group and control group by semi-quantitative RT-PCR.Result The level of Kiss-1 mRNA expression in the glandular cells in adenomyosis group was obviously decreased compared with control group during both proliferative and secretory phase(P<0.05).Compared with adenomyosis group ,the expression of Kiss-1 mRNA was increased after intervention with mifepristone during both proliferative and secretory phase(P<0.05). The expression of Hpa mRNA in the glandular cells in adenomyosis group was obviously increased compared with control group during both proliferative and secretory phase(P<0.05), which was decreased in mifepristone group during both proliferative and secretory phase(P<0.05). But the level of Kiss-1 mRNA and Hpa mRNA expressions in the glandular cells did not differ during secretory phase compared with proliferative phase in the same group(P>0.05).Conclusion1 The expressions of Kiss-1 mRNA and Hpa mRNA in the glandular cells are not effected by menstrual cycle. The low expression of Kiss-1 mRNA and high expression of Hpa mRNA in glandular cells of eutopic endometrium in adenomyosis are involved in the invasion of human adenomyosis.2 Mifepristone can increase the expression of Kiss-1 mRNA and decrease the expression of Hpa mRNA in glandular cells from eutopic endometrium in adenomyosis and reduce the invasive endometrium, which suggested a therapeutic role in adenomyosis.
