Investigation Of Children In Chongqing And The Impact Of IL-12 On Banlance Of Cord Blood's Th1/Th2

OBJECTIVE:Allergic rhinitis(AR)is a common disease of our department today,this illness is often associated with asthma,now more and more people have this disease ,especialy in the industrated country.the course is more complex ,still have not known.Some studies demonstrate that the reason maybe is surroudings,life style ,food custom,life pessure ,inherit and /or infection,in all of this,inherit and infection are very important.Allergic rhinitis is an abnormal immune reponse of body .The Th1 and the Th2 cells are inbalance,And the Th2 cells abruptly increased in mucosa of nasal. Some circumstances stimulate infant may arouse a protecting immunal reponse by stimulating Th1 cells.make Th1 cells and Th2 cells to a new balance ,and the syptom of allergic rhinitis will reduce.IL-12 is a protein of lymph cell secreting.It can set off NK cell,enhance lymph prime cell by PHA inducing;Accelerate CD4+TH0 cell to Th1 cell,stimulate IFN-γ,TNF-αand IL-2,suppress Th2 cell secrete IL-4,IL-5 and IL-10;May suppress IgE secreting by IL-4. This study is an epidemic survey of students in elementary/primary school in Chongqing.then we study 40 pregant wemen who are strong.we use IL-12 to regulate Th1 and Th2 cells in the infant's cord blood .Then we would learn that IL-12,as a kind of cytokine,can regulate Th1 and Th2 cells,supress the allergic reponse set off,so it can prove that IL-12 may cure allergic rhinitis in clinic.METHODS:1. We use paper investigation ,look into 720 primary school students ,whether they have symtom of allergic rhinitis.2. We use paper investigation,look into 40 pregent wemen whose body is strong,we randomly divid the 40 case into 4 group:group control,group parents positive(group 1),group mather positive(group 2),group father positive(group 3),we extract the cord blood plasma and analyse the expression of IL-4,IL-10,TNF-α,IFN-γby enzyme-linked immunosorbent assay(ELISA).3. We extract cord blood mononuclear cell,and find the best stimulating consistency of IL-12 by MTT.then we use the consistency of IL-12 to stimulate CBMC ,we culture the CBMC for 72 hours in 5% co2,37℃.then we collect the cultrue liquid and measure the expression of IL-4,IL-10,TNF-α,IFN-γby ELISA.RESULTS:1. The disease's morbidity of primary students in Chongqing is 28%.The new case is 202.2. The expression of Th2 type cytokine in group1 and group 2 and group 3 is significantly increased . and the group 1 is the best one.3. We use IL-12 stimulate CBMC ,we find the expression of Th2 type cytokine declined. The expression of Th1 and Th2 is regulated.CONCLUSIONS:1. The survey show that the rate of allergic rhinitis has much well with inherit and surrouding .2. The expression of Th1,Th2 type cytokine of cord blood demonstrate that inherit is very important,and if the parents both have allergic diseases,their child will has more chance to get allergic diseades.3. We find IL-12 can regulate the expression of Th1/Th2 type cells ,it can decrease the consistency of Th2 type cytokine and increase the consistency of Th1 type cytokine.then IL-12 will have chance to alleviate the symtom of allergic rhinitis.
