Experimental Study Of Xiaodouning Cream On Treatment Of Acne

Objective:To determine the main contents metronidazole and clindamycin of the Xiaocuoning cream by HPLC and establish quality control method,study the pharmacodynamics and observe its toxicity.Methods:1.To determine the contents of the Xiaocuoning cream by HPLC:the mobile phase was 0.02 mol/L ammonium dihydrogen phosphate solution(pH3.0)and acetonitrile at the ratios of 70:30,Detection wavelength was UV at 210 nm,flow speed at 1.0ml/min,the column temperature was 40℃,and go with accelerated test and long-term test.2 Antibacterial test The minimal inhibitory concentration(MIC) and the minimal bactericidal concentration(MBC)of the Xiaocuoning cream against staphlocococus,Staphylococcus albus and propionbacteria were measured in vitro by means of liquid-cuobling dilution.3.The influence on the extent of ear swelling The ear swelling model was caused by smearing of croton oil on ear inner surface in mice.The Xiaocuoning cream was smeared on ear outside surface.The extent of ear swelling was measured.4.The influence on experimental ache keratinous model According to Kligma's method,coal tar was applied daily to the area nearby the opening of the rabbit ear canal to make keratinous acne model.The keratinous acne model were observed by naked eye and pathological histoligical examination in different groups.5.The toxicity test The Xiaocuoning cream was smeared on the rabbit skin for the skin irritating test and the acute toxicity test on in tact and broken skin respectively,the Xiaocuoning cream was smeared to pig for the skin allergic test.Results:1.The HPLC method had a good selectivity and reproducibility while the adjuvants could be separated from the main compositions.The stability study on preparation indicated that it was stable,nearly without any changes during the stability study.2.Antibacterial test the Xiaocuoning cream had obvious effects on staphylocococus,Staphylococcus albus and propionbacteria.3.The influence on the extent of ear swelling Different doses of the Xiaocuoning cream have depression to ear swelling extent in mice.Compared with the normal saline group and base group,significant difference was showed.4.The influence on experimental keratinous acne model The ear keratinous acne model was formed successfully in the rabbits.In the Xiaocuoning cream treated groups,the tissue pathology of rabbits' ears showed that inflammation lightened obviously, fair folliclus keratinous plugs decreased and disappeared,and the debouches of fair folliculus became flat compared to the base group.5.The toxicity test The Xiaocuoning cream has no irritability,allergia and toxicitiy.Conclusions:The method of quality control is simple,rapid and accurate;it has notable effects on bacteiostatic and anti-inflammatory effects,and is safe and effective for exteral use.
