The Recipe of Acne Cream is an salted recipe which have been used for the therapeutics to ache in department of dermatology of XiYuan Hospital for a long time.It has been proved that this recipe has great effect to clinical treatment for acne and set a good foundation for the further study.Ache Cream is a new compound preparation of Traditional Chinese Medicine which consists of Radix et Rhizoma Rhei,Hedyotic diffusa,Folium Isatidis,Herba Artemisiae Scopariae,Rhizoma Atracylodis,Radix Salviae Miltiorrhiae,Raidix Sanguisorbae,Radix Glycytthizae,Broneolum Syntheticum.It has the function of clearing away Heat-toxin, eliminating dampness and cooling the blood.It can be used to cure acne which caused by dampness and heat invading internal,turbid invasion,dampness-heat and blood stasis blocks to attack face and induce to Ache.With the guidance of the theories of Traditional Chinese Medicine,Acne Cream was studied with modern pharmaceutical technology.For the demand of clinic,the form of drug was chosen as ointment.On the basis of the function and treatment as well as the character of the component of the herbs,the extract conditions were selected by using orthogonal design: Decoct all the Chinese crude drug except Broneolum Syntheticum with water 12 times,1.5 hours for each time,decoct 3 times.Combine the decoctions and filter,concentrate the filtrate,add 95%ethanol to produce a solution containing 65%of ethanol,stir well,standing 24 hours,filter,recover ethanol.put it into vacuum drying oven until it become powder for later use.Add above powder to water substrate and add Broneolum Syntheticum to oil substrate.,Raise the temperature and combine water phase and oil phase and use agent-in-oil method to form the ointment.By means of selecting fight substrates and optimizing the quantity of emulsification agent, water phase bases and oil phase bases,we prepared O/W cream which was stable and had good appearance and properties.After that the quality standard of Ache Cream was finished.Determine the content of emodin and chrysophanol in Acne Cream by HPLC.The mobile phase consisted of methanol: 0.1%Phosphoric Acid Solution(85:15).The detected wavelength was set at 440nm.It is resulted that the linearity of this method was good.In the range of emodin was 0.024～0.3μg(r=0.9996);chrysophanol was 0.02～0.25μg(r=O.9996).The recoveries of emodin and chrysophanol were 99.5%and 97.4%,RSD were 2.98%and 2.33%,respectively.The method is accurate,precise and is suitable to detect emodin and chrysophanol in Acne Cream..We also study skin permeation test to Acne Cream.It were collected by using Franz diffusion cells after permeation through the skin of rats.The content of emodin and chrysophanol in Acne Cream were determined by HPLC.Under the condition of an effective area of 3.80cm~2,it was found that the result was stable and the reproducibility was well with drug loading more than 0.5g,the receiver solution ethanol to normal saline=5:5(v:v).The standardization of Chinese Medicine Cream percutaneous test in vitro was discussed preliminarily.To study the irritation of Acne Cream,we applied hair-cut healthy SD rats with Acne Cream one or more times.No irritation reaction occurred in complete skin of SD rats.The result indicated that Acne Cream was safe as topical agent.To sum up,after synthetically assessment of Acne Cream,we can say that it is a safe, effective and stable topical agent for the treatment of Acne and have good marketing promise deserving to popularize.
