The Study On The Therapeutic Mechanisms Of The Prescription Warming The Kidney And Invigorating The Spleenon Rat's Experimental Hypothyroidism

Objective:Investigate the therapeutic effects of the prescription warming the kidney andinvigorating the spleen on the rat's model of the hypothyroid deficiency, and it'stherapeutic mechanisms. They were divided into 4 groups by random(10 per group): the model group, the prescription wanning the kidney and invigoratingthe spleen (the traditional Chinese medicine group), the Western medicinegroup(Thyroid Tablets group), and the contrast group. The model group and thetherapeutic groups were poured 0.1ï¼…PTU fluid into stomach one time each day. Thecontrast group was poured the same quality of tap water. 15 days later, we sampledthe blood from every rat's angulus oculi to check the thyroid function, and confirmedthat the model had been made. After that, based on administering PTU, we began tocure the rats. We administered the 0.08ï¼…Thyroid Tablets fluid to the Western medicinegroup, 0.9ml/100g body weight (about 0.72g/100g crude drug) the self-made herbalmedicine fluid to the traditional Chinese medicine group,. In the remedial process, weobserved the rat's appearance, spirit, behavior, dejecta, food-intake, rectaltemperature, weight and heart rate.8 weeks later, we executed all the rats andsampled the blood to check the thyroid function, E₂,ACTH, Cot, SOD, IgG, RBC, Hb,ALT, AST, BUN and Cr. Then we anatomized the rats, and separated the half ofthyroid glands to check the tissular SOD. We select the pituitaries, the another half ofthyroid glands, the livers, the kidneys, and the adrenal glands to check theirhistopathological change.Results:1. The symptoms and the objective signs of the therapeutic groups had improved ina certain extent. The food-intake had increased. The rectal temperature and theheart rate had risen.2. The TH of the model group was lower than the contrast group's(P＜0.05). The TSH of themodel group was higher than the contrast group's(P＜0.05). The FT₄ and T₄ of the therapeuticgroups had no change(P＞0.05). The FT3 of the the traditional Chinese medicine groups hadrisen(P＜0.01). The T3 of the traditional Chinese medicine groups had risen(P＜0.05). TheTSH of the traditional Chinese medicine groups was lower than the contrastgroup's(P＜0.05), but was also abnormal. The TSH of the Western medicine group had beennormal. 3. The Cor of all the rats had no change. The ACTH of the model group was higherthan the contrast group's(P＜0.05). The ACTH of the high dosage of the traditionalChinese medicine group had been normal. The E₂ of the model group was lowerthan the contrast group's(P＜0.05). Compared with the model group,the E₂ of allthe therapeutic groups had risen(P＜0.05).4. The serumal IgG of the model group was higher than the contrastgroup's(P＜0.01). The IgG of the traditional Chinese medicine groups were similarwith the model group's(P＞0.05).5. The serumal SOD of the model group was lower than the contrastgroup's(P＜0.01). The contrast group had no difference with., and the traditionalChinese medicine groups (p＞0.05). The serumal SOD had the positive correlationwith the T₃,the T₄,the FT₃, the FT₄(P＜0.05), and the negative correlation with theTSH (P＜0.05). The tissular SOD of the model group was higher than the contrastgroup's(P＜0.01).6. The RBC and the Hb of the model group were markedly lower than the contrastgroup's(P＜0.01). The traditional Chinese medicine group could increase the RBCand the Hb.(p＜0.05). The Western medicine group was similar with the modelgroup(p＞0.05)and was lower than the contrast group (p＜0.01), It hints that Hb ofthe Western medicine group cut down significant.7. The ALT of the model group was higher than the contrast group's(P＜0.01). All thetherapeutic groups'ALT had been normal. This experiment had no effect on theAST and the BUN. The Cr of the model group was higher than the contrastgroup's(P＜0.01). The Cr of the traditional Chinese medicine group and theWestem medicine group had been normal.8. The pathological check showed that the rats' thyroid glands were obviouslyintumescent. Under the light microscope, we found that the thyroid glands hadthe compensatory hyperplasia; the basophils in the anterior pituitary hadincreased, the kytoplasm had a lot of basophilic stippling; the cells of the adrenalcortex had mild hyperplasia. All the organ of the Chinese therapeutic groups hadsome amelioration. The livers and the kidneys had no obviously pathologicalchange, the model group had no difference with the contrast group.Conclusions:The prescription warming the kidney and invigorating the spleen, may inspire yang-qito promote the functional activity of the body, and it has good therapeutic effect onhypothyroidism rats, It can ameliorate the symptoms and the objective signsobviously, It can promote the TH, reduce the sTSH. It can adjust the disorder of thepituitary gland-adrenal gland and the pituitary gland-genitical gland. It can reducethe exorbitant serumal IgG, relieve the harm by free radical. It can promote the RBCand Hb, ameliorate the hypothyroid anemia. It also can reduce the ALT.
