The Study On The Therapeutic Effect Of You Gui Pills On Rat's Experimental Hypothyroidism And Its Therapeutic Mechanisms

Objective:Investigate the therapeutic effects of the You Gui pills on the rat's model of the hypothyroid deficiency of the spleen-yang and the kidney-yang ,and it's therapeutic mechanisms .This is good to excavate ancient prescription ,and to provide a new idea and some experimental evidence in order to apply it in clinic better. Methods:1. This experiment used 70 female SD rats .They were divided into 7 groups by random :the model group ,the low dosage of You Gui pills group , the middle dosage of You Gui pills group , the high dosage of You Gui pills group ,the Thyroid Tablets group ,the You Gui pills with Thyroid Tablets group,and the contrast group . The model group and the therapeutic groups were poured 0.1% PTU fluid into stomach one time each day .The contrast group was poured the same quality of tap water .15 days later ,we sampled the blood from every rat's angulus oculi to check the thyroid function ,and confirmed that the model had been made.2. After that ,based on administering PTU ,we began to cure the rats .We administered the 0.08% Thyroid Tablets fluid to the Thyroid Tablets group ,0.140g/ml You Gui pills fluid to the low dosage of You Gui pills group ,0.275g/ml You Gui pills fluid to the middle dosage of You Gui pills group ,0.550g/ml You Gui pills fluid to the high dosage of You Gui pills group , 0.275g/ml You Gui pills fluid with 0.08% Thyroid Tablets fluid to the You Gui pills with Thyroid Tablets group.3. In the remedial process ,we observed the rat's appearance ,spirit ,behavior , dejecta, food-intake, rectal temperature , weight and heart rate .8 weeks later ,we executed all the rats and sampled the blood to check the thyroid function ,E2,ACTH,Cor,SOD,IgG,RBC,Hb,ALT,AST,ALP,BUN and Cr .Then we anatomized the rats , and separated the half of thyroid glands to check the tissular SOD .We select the pituitarys ,the another half of thyroid glands ,the livers ,the kidneys ,and the adrenal glands to check their histopathological change.4. We analyzed the data by analysis of variance ,q-test and analysis of linear correlation.Results:1. The symptoms and the objective signs of the therapeutic groups had improved in a certain extent. The food-intake had increased .The rectal temperature and the heart rate had risen.2. The TH of the model group was lower than the contrast group's(P<0.05) .The TSH of the model group was higher than the contrast group 's(P<0.05) .The FT4 and T4 of the therapeutic groups had no change(P>0.05) .The FT3 of the You Gui pills groups and the You Gui pills with Thyroid Tablets group had risen(P<0.01). The T3 of the You Gui pills groups had risen(P<0.01). The TSH of the Thyroid Tablets group and the You Gui pills with Thyroid Tablets group had been normal.3. The Cor of all the rats had no change .The ACTH of the model group was higher than the contrast group's(P<0.05) .The ACTH of the high dosage of You Gui pills group and the You Gui pills with Thyroid Tablets group had been normal .The E2 of the model group was lower than the contrast group 5s(P<0.05). Compared with the model group ,the E2 of all the therapeutic groups had risen(P<0.05).4. The serumal IgG of the model group was higher than the contrast group 's(P<0.05) .The IgG of the therapeutic groups were similar with the model group's(P>0.05).5. The serumal SOD of the model group was lower than the contrast group's(P<0.01). The Thyroid Tablets group had no difference with the contrast group .The serumal SOD had the positive correlation with the T3 ,the T4 ,the FT3 ,the FT4(P<0.05) ,and the negative correlation with the TSH (P<0.05) . The tissular SOD of the model group was higher than the contrast group5s(P<0.01).6. The RBC and the Hb of the model group were lower than the contrast group 's(P<0.01) . The You Gui pills with Thyroid Tablets could increase the RBC and the Hb.7. The ALP of the model group was higher than the contrast group 's(P<0.01) . The ALP of the middle dosage of You Gui pills group had been normal. The ALT of the model group was higher than the contrast group's(P<0.01) .All the therapeutic groups'ALT had been normal. This experiment had no effect on the AST and the BUN . The Cr of the model group was higher than the contrast group 's(P<0.01) . The middle dosage of You Gui pills group ,the high dosage of You Gui pills group ,the Thyroid Tablets group and the You Gui pills with Thyroid Tablets group had been normal.8. The pathological check showed that the rats' thyroid glands were obviouslyintumescent . Under the light microscope ,we found that the thyroid glands had the compensatory hyperplasia;the basophils in the anterior pituitary had increased ,the kytoplasm had a lot of basophilic stippling;the cells of the adrenal cortex had mild hyperplasia .All the organ of the Chinese therapeutic groups had some amelioration .The livers and the kidneys had no obviously pathological change ,the model group had no difference with the contrast group . Conclusions:The therapeutic mechanisms of the You Gui pills to the hypothyroid rats is different from the TH's .It can ameliorate the symptoms and the objective signs obviously ,and this effect is better than Thyroid Tablets only .It can promote the TH, reduce the sTSH .It can adjust the disorder of the pituitary gland- adrenal gland and the pituitary gland- genitical gland .It can reduce the exorbitant serumal IgG, relieve the harm by free radical .It can promote the RBC and Hb , ameliorate the hypothyroid anemia .It also can reduce the ALP ,and this may relate to that it could adjust the imbalance of bone's metabolism due to the hypothyroidism . In the histology ,the You Gui pills can relieve the compensatory hyperplasia of the thyroid gland ^meliorate the excessive incretion of the basophils;adjust the pathological changes of the adrenal gland . We had not found the You Gui pill had adverse drug reactions to the liver and the kidney .It can protect the injured liver and kidney ,so it is safe and credible.
