The Influence Of Acupuncture Combined With Herbal Medicine On Angiogenesis (VEGF,bFGF) In Rat Mammary Hyperplasia Model

Objective: Mammary hyperplasia as a benign lesion , is a common illness and a frequently encountered disease for child-bearing-period women . Its incidence rate accounts for 74.1% of all the breast diseases , and it has an ascendant tendency year by year .The crowd with the disease have 2~3 times incidence than the common . So , active treatment in mammary hyperplasia has much important significance for preventing its canceration , and also can raise quality of life of patients . To existenting data , morden medicine is still lack of satisfactory therapeutic measure on the disease . Its endocrine therapy has much adverse reaction , long-time use may interfere the endocrine balance . Therefore , it is not suitable as routine treatment . Our study made use of two therapeutic tools in Chinese traditional medicine--inese crude drug and acupuncture , from the view of angiogenesis , approached the mechanism of action of acupuncture combined with herbal medicine on mammary hyperplasia .We duplicated rat models of mammary hyperplasia ,adopted three treatments : simplely herbal medicine , simplely acupuncture and western medicine sanbenyangan-capsule(TAM) , observed their curative effect ; and also studyed the changes of some genital hormones and angiogenesis factors . It will abundant and develop practitioner of Chinese medicine .Method: 60 depuratory and healthy Wistar female rats , 75-85-day in age, 220±20g in bodyweight , were divide into 6 groups randomly : Normal group, Model group, Control group , Acupuncture group , Herbal medicine group , Acupuncture combined with Herbal medicine group . There were 10 rats in each group . Rats in normal group were raised by routine feed , the other five groups were duplicated the model of mammary hyperplasia . Based on the method of Zhanglu: these rats were injected estradial benzoate , in dose of 0.5 mg/kg , once a day for 25 days , then injected progestin , in dose of 5 mg /kg , once a day for 5 days . From the thirty-first day , each group adopted group therapy . Acupuncture group:After modeling , We chose the acupucture points of"wuyi""hegu""tanzhong""zusanli", once a day , nine days as a course of treatment , for three courses . Herbal medicine group : after modeling , rats were administrated xiaozhengsanjietang-herb , in dose of 25g/kg , once a day for 30 days . Acupuncture combined with Herbal medicine group : after modeling , acupucture points were the same as acupuncture group, the medicine doses were the same as herbal medicine group , for 30 days . Control group: after modeling , rats were given TAM , in dose of 1.8 mg/kg , the same as clinical equivalent dose , once a day for 30 days . The rats in Normal group and Model group were given the same voluminal distilled water for 30 days . Fasting 5 hours after last administration, measured the second mammary diameter with sliding caliper , collected blood , separated serum , placed them respectively , detected the serum levels of estradiol (E2) , progestogen (P) and prolactin (PRL) by means of radioimmunology . Cut rats mammary quickly , slightly wiped with filter paper , fixed mammary in 4% polyoxymethylene , paraffin embedded , slice in 3 um . HE dyeing , observed the mammary histomorphology , detected the expression of VEGF and bFGF in mammary glandular tissue by euzymelinked immunosorbent assay .Result:1 Changes of mammary diameterCompaired with normal group , the mammary diameters in model group were bigger , and there were significant difference between two groups (P<0.05). After therapy , mammary diameters in Acupuncture group and Herbal medicine group were shorter than that in Model group , the difference was of statistical significance(P<0.05) . The mammary diameters in Acupuncture combined with Herbal medicine group and Control group , Compaired with Model group , were much shorter , and there were significant difference(P<0.01) .2 Changes of genital hormone in serumCompaired with normal group , the contents of serum E2 in model group were markelly increased , and there were significant difference between two groups (P<0.01). After therapy , contents in Acupuncture group and Herbal medicine group were decreased , and the difference was of statistical significance (P<0.05). The contents in Acupuncture combined with Herbal medicine group and Control group , Compaired with Model group , were markelly decreased , and there were significant difference(P<0.01).Compaired with normal group , the contents of serum P in model group were markelly decreased , and there were significant difference between two groups(P < 0.01). After therapy , Control group, Acupuncture group and Herbal medicine group all had an increasing tendency , but it was of no statistical significance(P>0.05) . Its contents in Acupuncture combined with Herbal medicine group, Compaired with Model group , were markelly increased , and there was significant difference (P<0.01).Compaired with normal group , the contents of serum PRL in model group were markelly increased , and there were significant difference between two groups(P < 0.05). After therapy , Control group, Acupuncture group and Herbal medicine group all had a decreasing tendency , but it was of no statistical significance(P>0.05) . Its contents in Acupuncture combined with Herbal medicine group, Compaired with Model group , were markelly decreased , and there were significant difference(P<0.05).3 Changes of VEGF and bFGF in serumCompaired with normal group , the level of serum VEGF in model group was markelly increased , and there was significant difference between two groups(P < 0.01). After therapy , there were decreasing tendency in Acupuncture group and Herbal medicine group , but it was of no statistical significance (P>0.05). The level of serum VEGF in Control group was lower than that in Model group , the difference was of statistical significance (P<0.05). The level of serum VEGF in Acupuncture combined with Herbal medicine group were markelly lower , Compaired with Model group , and there was significant difference (P<0.01).Compaired with normal group , the level of serum bFGF in model group was markelly increased , and there was significant difference between two groups(P<0.01). After therapy , there was a decreasing tendency in Acupuncture group , but it was of no statistical significance (P>0.05). The level of serum bFGF in Herbal medicine group was lower than that in Model group , the difference was of statistical significance(P<0.05) . The level of serum bFGF in Acupuncture combined with Herbal medicine group and Control group were markelly lower , Compaired with Model group , and there were significant difference(P<0.01).4 Expression of VEGF and bFGF in mammary glandular tissue Positive expression of VEGF in mammary glandular tissue were located in intracytoplasm, stained dark orange . Compaired with normal group , there was significantly higher expression of VEGF in model group(P<0.01). After therapy , there was a decreasing tendency in Control group , but it was of no statistical significance(P>0.05) . The expression of VEGF in Acupuncture group , Herbal medicine group and Acupuncture combined with Herbal medicine group were markelly lower , Compaired with Model group , and there was significant difference(P<0.01).Positive expression of bFGF in mammary glandular tissue were located in intracytoplasm , stained dark orange. Compaired with normal group , there was significantly higher expression of bFGF in model group(P<0.01) . After therapy , there was a decreasing tendency in Control group, but it was of no statistical significance(P>0.05). The expression of bFGF in Acupuncture group , Herbal medicine group and Acupuncture combined with Herbal medicine group were markelly lower , Compaired with Model group , and there was significant difference (P<0.01).Conclusion:1 Compaired with normal group , the contents of serum E2 and PRL in rats model of mammary hyperplasia were markelly increased , the contents of serum P was lower . It told us that endocrine disturbance was the important factor of mammary hyperplasia.2 The level of serum VEGF and bFGF in model group , compaired with normal group , was markelly increased , and they had higher expression in some mammary gland tissue . It indicated that some angiogenesis factors were actived to induce angiogenesis directly or indirectly . 3 Acupuncture combined with herbal medicine on mammary hyperplasia , can adjust some genital hormones , decresed the contants and the expression of some angiogenesis factors , and also improved endocrine secretion , improved microcirculation,to achieve curing mammary hyperplasia.
