Radix Gentiana is one of the commonly used traditional Chinese medicines (TCM). 40 Radix Gentiana samples were collected from different habitats and breeds and the quality assessment was developed.A RP-HPLC method was developed for determination of gentiopicroside (GTP) in Radix Gentiana. A Diamonsil ODS column was used with the mobile phase being acetonitrile-water-acetic acid (10:80:l,v/v), the internal standard was p— hydroxybenzoid acid and the detection wavelength was set at 270 nm. The calibration curve was linear (r=0.9999) in the range of 12.8-153.6 μg·mL-1 for gentiopicroside, the average recovery of the method was 99.1%(RSD =0.4%). A RP-HPLC method was established for determination of swertiamarin (SWT) and sweroside (SWO) in Radix Gentiana. A Diamonsil ODS column was used, the mobile phase was acetonitrile-water-acetic acid (10:105:l,v/v) and the detection wavelength was set at 240 nm. The linear response ranges were 4.0 64 μg·mL-1 for swertiamarin (r=0.9999), 3.2 96 μg·mL-1 for sweroside (r=0.9998). The average recoveries of swertiamarin and sweroside were 97.7% (RSD=1.4%) and 97.9% (RSD=1.2%) respectively.
