Objective: Intraperitoneally injecting the1,2,3,4,6-penta-O-galloyl-?-D-glucose(?-PGG) into diabetic rats and normal rats to observe the 1,2,3,4,6-penta-O-galloy l-?-D-glucose concentration changes in the pancreas and analyze its effect to TXNIP and Trx1 protein expression. Method:the 1,2,3,4,6-penta-O-galloyl-?-D-glucose(?-PGG) into diabetic rats and normal rats;To measure the concentration changes for the Gallic acid in the blood and pancreas of the diabetic and normal rats by using HPLC with Intraperitoneally injection; to measure the TXNIP and Trx1 protein expression in the pncreas tissue of diabetic rats and normal rats by Western blot method. Result: ?-PGG is intraperitoneally injected once and continuously respectively. The Gallic acid is found in the pancreas but not in the blood of both diabetic rats and normal rats by HPLC method. Moreover, the rates of concentration of Gallic acid in the diabetic rat group are higher than those of the normal rat group, which has the statistical difference(P<0.05). The result of measuring the TXNIP and Trx1 protein expression by Western blot method: the TXNIP protein expression in the model group is higher than that in the control group, which has the statistical difference(P<0.05); the Trx1 protein expression in the model group is lower than that in the nomal group, which has the statistical difference(P<0.05). Conclusion:The Gallic acid is found in the pancreas of both diabetic rats and normal rats by HPLC intraperitoneal injection. Moreover, the concentration of Gallic acid decomposed from pancreas of the diabetic rat group is higher than that in the normal rat group with the same ?-PGG concentration injection, which has the statistical difference(P<0.05). There is the statistical difference(P<0.05), to compare the TXNIP and Trx1 protein expression in the model group and the control group by Western blot method; in addition, in the pancreas of the diabetic rat group, with the increase of?-PGG concentration, Trx1 protain expression also increases. Suggestion: ?-PGG could have beneficial effect on survival of pancreatic?-cell.
