The disease incidence of breast cancer has trend of increasing. In some areas, especially in some big cities, it has leaped to first place among the malignant tumors. Its ten-year survival rate is approximate 40 percent. But, through the everyday clinical practices, people have found that the patients whose traditional prognosis predictor situation is resembled don't have resembled outcome. Then people believe that only depend on the traditional prognosis predictor will influence the judgment of many patients' prognosis. At present, the researches about breast cancer prognosis related biological predictor have become a hot spot in the field. There are many researches have proved that the over-expression of C-erbB-2, a kind of oncogene, is an important factor related to the poor prognosis of the breast cancer, and PCNA is an important sign about the ability of the tumor cell proliferating. The two predictors are all closely related to the breast cancer prognosis Syk is a new sign about the breast cancer prognosis, which is put forward in recent years. Now days, there are few research about Syk. The relationship between Syk and the traditional prognosis predictors and other breast cancer prognosis predictors are not clear. In this study, we examined the expression of C-erbB-2,PCNA and Syk in human breast cancer tissue by using immnohistochemical staining, to check the relationship between Syk and C-erbB-2,PCNA and the traditional prognosis predictors and the outcome of five-year survival rate, then elucidate the value of expression of Syk in breast cancer prognosis and treatment. Methods: The expression of C-erbB-2,PCNA and Syk were examined by immunohistochemistry in 5 specimens of normal breast tissue, 5 specimens of breast fibroadenomas and 35 specimens of primary breast cancer (The patients are all female from 20-68 years old. We had followed-up the patients 60 months and found 9 patients has died in 5 years. The all patients didn't have chemotherapy or radiotherapy before operation therapy. There are 19 patients whose auxiliary lymph nodes are positive, and 16 patients whose auxiliary lymph nodes are negative.). The 35 specimens of primary breast cancer are divided into a certain number groups on the basis of the result of five-year-survival, clinical stages, the size of tumor, etc. The staining result was analyzed by using the optical microscope, then investigating the difference between corresponding groups.Results: In breast fibroadenomas and normal breast tissue, we have found the C-erbB-2 and PCNA are negatively expressed in all specimens, while Syk is positively expressed in all specimens.C-erbB-2 is expressed mainly in carcinomatous cytoplasm of breast cancer. The positive reaction appears brightly brown-yellow staining. The positive ratio of C-erbB-2 expression is 51.4%(18/35) in the 35 breast cancer specimens. The positive ratio of C-erbB-2 in the patients who survived over five years is 38.5%(10/26) is lower than those who lived less than five years (88.9%, 8/9). There is statistical difference between them (P<0.05).PCNA is expressed mainly in cell nuclear of breast cancer. The positive reaction appears brightly brown-yellow staining. The positive ratio of PCNA expression is 68.6%(24/35) in the 35 breast cancer specimens. The positive ratio of PCNA in the patients who survived over five years is 57.7%(15/26) is lower than those who lived less than five years (100%, 9/9). There is statistical difference between them (P<0.05).Syk is expressed mainly in carcinomatous cytoplasm of breast cancer. The positive reaction appears brightly brown-yellow staining. The negative ratio of Syk expression is 31.4%(11/35) in the 35 breast cancer specimens. We have found that there is significant correlation between the expression of Syk and PCNA (P<0.05), but there isn't significant correlation between the expression of Syk and C-erbB-2 (P>0.05).
