Objective To analysis the expression of C-erbB-2, P53, estrogen receptor (ER), progesterone receptor(PR), proliferating cell nuclear antigen(PCNA), ki67 protein and Cyclooxygenase 2 (Cyclooxygenase, COX2) in breast cancer, and clinical pathology, and significance of the co-expression. Method 20 paraffin cases of breast adenosis and 60 frozen or paraffin cases of breast cancer were collected, from February 2007 to February 2009, in Department of Pathology, Central Hospital, Siping paraffin specimens. Among 60 cases of breast cancer, only one was male and the rest were female, age from 31 years to 81 years, the average age--52.93±9.99; 53 cases of infiltrating ductal carcinoma,7 cases of Non-invasive ductal carcinoma (that is ductal carcinoma in situ). SP immunohistochemical staining was used to test the expression of P53 (DO7), C-erbB-2 (CB11), ER (ID5), PR (SP2), Ki67 (SP6), PCNA (PC10), COX2 (COX SP21) in breast cancer and breast cancer tissue adenosis.Resultl. The positive expression of P53, C-erbB-2, ER, PR, ki67, PCNA, COX in Breast cancer was significantly higher than the patients with breast adenosis. The positive expression rate:P53 60.00%(36/60), C-erbB-238.33%(23/60), ER 68.33% (41/60), PR 61.67%(37/60), ki67 66.67%(40/60), PCNA 78.33%(47/60), COX2 56.67%(34/60). There was significantly difference (P<0.05).2. The positive rate of P53 expression in breast cancer patients (60%) was significantly higher than the C-erbB-2 (38.3%) (x2=5.63, P< 0.05).3. Breast cancer P53, C-erbB-2 expression had a higher rate of lymph node metastasis (80.8% and 65.4%) (P< 0.01).But the positive expression of ER, PR, ki67, PCNA, COX2 and lymph node metastasis was no significant difference.(P> 0.05).4. The P53, C-erbB-2, ER, PR, ki67, PCNA, COX2 in Breast cancer had no correlation with age, tumor size, clinical stage. (P> 0.05) 5. All histological types of breast cancer sample and P53, C-erbB-2, ER, PR, ki67, PCNA, COX2 expression was no significant difference (P value> 0.05).Conclusion1. The P53 and C-erbB-2 in breast cancer patients was significantly higher than the patients with benign breast adenosis. The protein expression of P53 and C-erbB-2 gene in breast cancer patients had a higher rate of lymph node metastasis than ER, PR, ki67, PCNA, COX2 detection index. That proved P53 and C-erbB-2 had significant difference in breast cancer development and metastasis. They also can be indicators of the prognosis of breast cancer.2. P53 and C-erbB-2, ER, PR, ki67, PCNA, COX2 expression was no significant difference with age, tumor size, clinical stage, pathologic type.3. The positive expression of P53, C-erbB-2, ER, PR, ki67, PCNA, COX2 was both independent and interrelated and was used as the reference drug in breast cancer patients. Combined detection of P53, C-erbB-2 and ER, PR, ki67, PCNA, COX2 gene can be used as the evaluation of the development of breast cancer.
