Objective: In this study, we want to investigate the c-erbB-2, P53 and PCNA expression in breast cancer and to reveal the correlation between their expression and clinical-pathological parameters of breast cancer and to analyze their function in the occurrence and progression of breast cancer. To establish the real time RT-PCR method to measure the expression of c-erbB-2 mRNA in breast cancer tissues of beside tumors and tissues of adenosis of mammary glands, to find out the relationship of c-erbB-2 protein with its mRNA and make out their clinical values respectively.Methods: 77 specimens of breast cancer and 77 specimens of beside tumors were evaluated by IHC technique with antibodies against c-erbB-2, P53 and PCNA. The real time RT-PCR method for detecting the expression of c-erbB-2 was established successfully.38 specimens of breast cancer , 17 of beside tumors, 10 of adenosis of mammary glands were detected in the Lightcyler Applied Biosystem Sequence Detector.Results: Expression of the c-erbB-2, P53 and PCNA was detected in 39.0%, 48.1%,85.7% of carcinomas. There is correlation of the c-erbB-2, P53 and PCNA and the clinical-pathological parameters of breast cancer. c-erbB-2 has positively correlation with P53, rS =o.244, P<0.05.There is significant correlation between P53 and PCNA, rS =0.393, P<0.05. c-erbB-2 has no correlation with PCNA .There is significant difference in the level of c-erbB-2 mRNA among the three groups(P<0.05). The level of c-erbB-2 mRNA has correlation with the C-erbB-2 protein (P<0.05).Conclusions: The expression of c-erbB-2, P53 and PCNA is elevated in breast cancer, Detecting them will assist clinical doctors in judging the biological behavior of tumor and directing therapy.
