Objective: To study the associations between plasma fibrinogen level of ischemic stroke and genevariation(-455G/A)Located at -455 bp from the start of transcription of the -figrinogen gene . Method : 86 hypertensive patients with ischemic stroke (stroke group) . 85 hypertensive patients without ischemic stroke (hypertensive group) and 90 healthy blood donors (control group) were randomly enrolled in this trial . p-fibrinogen gene -455G/A polymorphy was analyzed from all subjects by polymerase chain reaction with restrictive enzyme HaeIII(PCR-RFLP method) . Genomic DNA was extracted from white blood cells using salt/chloroform method. Plasma fibrinogen levels were measured with prothrombin time (PT) assasy . Result The frequency of the A-455 allele in control group was 0.11 (95%CI:0.05~ 0.17) . The frequency of the A-455 allele in the hypertensive group and strok group were 0.13 (95%CI:0.06-0.20) and 0.22 (95%CI:0.13~0.31) .A'455 allele frequency of the stroke group was significantly higher than that of control group (x2=7.01, P<0.01) and the hypertensivegroup (x2 = 4.40, P<0.05) . The mean SD fibrinogen level was significantly higher in the stroke group than that in the control group . (4.82 0.26g/L versus 4.37 0.19g/L,t=2.79, P<0.01) , while in hypertensive group the differance was not significant compared with both stroke group and control group . Plasma fibrinogen levels in patients with -455G/G gene were signiticantly lower than that with -455G/A and -455A/A gene in both stroke group and hypertensive group , but the differance was not in the control group. Conclusion Plasma fibrinogen level is affected by the p-fibrinogen gene -455G/A polymorphism . The fibrinogen A-455 allele may be an independent risk factor for ischemic stroke.
