Objective: To analyse the expression and distribution of pKi67 in hypertrophic scar and keloid tissue, identify the characteristics of cell proliferation, and find an efficient diagnostic method of the two types of pathological scars.Methods: Immunohistochemistry(SABC method) was employed to detect the expression and distribution of pKi67 in keloids, hypertrophic scars with rabbit-anti-human polycolonal Ki67 antibody. Biopsies were taken from 21 cases of hypertrophic scar patients, 19 cases of keloid patients, and 8 cases of healthy people with normal skin. Image analysis was performed to quantitatively compare the differences of pKi67 expression in these tissues. Results: Nucleuses were positively stained both in fibroblasts of dermis and some keratinocytes of epidermis including basel layer, stratum spinosum, and part of granular layer. The pKi67IOD(integrated optical density) value in keloids (401.82±114.29)was significantly higher than that in hypertrophic scars(328.21 ± 131.40); the pKi67IOD value in hypertrophic scars was singnificantly higher than that in normal skins(213.92±66.62); P<0.05. The pKi67IOD value in hypertrophic scars was inversely correlated with its duration(r=-0.491, P<0.05) .Conclusion: Keratinocytes and flbroblasts in pathological scars kept high proliferating activity, may play an important role in scar formation. The characteriristic of flbroblasts in keloids differed from hypertrophic scars, which was probably due to their different subpopulation of flbroblasts.
