| PrefacePerfluoroethane (C2F6) is one of inert gases that have long influence. It was used in the treatment of the retinal detachment about 20 years ago. With the development of microscopic operation, more and more Ophthalmologists admit C2F6 as the substitute of vitreous body. Purulent endophthalmitis has such characteristics as spreading fiercely, damaging tissue seriously, bad prognosis and so on. The treatment for it includes two aspects: one is using antibiotic and steroid through all kinds of approaches, the other is the operation-vitrectomy.In this experiment, we observe the influence of C2F6 on purulent endophthalmitis through cultivating the bacteria in vitreous cavity and researching the pathologic retinal tissue slices through optic microscopes. Both the specimens needed come from the animal patterns.Materials1.Animals: 30 standard laboratorial albino rabbits with big ears, no limitation with sex.2.Equipment: upside-down microscope, olefin slicer etc.3.Medicine: inert gas Perfluoroethane (C2F6), SUMIANXIN, Gentamycin etc.Methods1.Execution for animal patterns: At about 1.5mm behind the limbus, inject vertically into vitreous cavity 0.05ml (about 30-50 Entries) straphylococcus aureus. After 10 to 16 hours, if there are pyic exudates in the papillary area or middle degree vitreous opaque that disturbs the observation of the second grade veins around optic papilla, the animal pattern of purulent endophthalmitis has been made successfully.2.Grouping: After numbering the animals (eyes), divide them into four groups. The eyes in No. 1 group are injected Gentamycin (10 eyes) into vitreous cavity, those in No.2 group are injected C2p6 (22 eyes). Injected C2F6+Gentamycin into the eyes in No.3 group (23 eyes) and sterilized air into those in No.4 group.3.Collecting of specimen: Give the animals check-up everyday by slit-lamp microscope, direct ophthalmoscope and non-contacttonometer and use antibiotic eye drops three times a day. Under general anesthesia, we extract vitreous body O.lml to cultivate bacteria at 7 days before and every 7 days after the execution of treatment.1.Methods of germiculture: Scatter the extracted vitreous body onto the agar culture media and then cultivate them in constant temperature boxes 24 hours, research, if find some bacteria growing, smear them, dye and distinguish the nosogenesis.2.The execution of the pathologic slices: Fix the eyeball onto the worktable and then cut a triangle incision in the center of cornea (each side of triangle is about 2mm) for the Formalin can act thoroughly. 72 hours later, draw materials, dehydrate, embed with olefm, slice, HE dyeing and seal up the glass slides.3.Methods of observation: Purulent endophthalmitis has a lot of research targets; we mainly evaluate the situation of vitreous opaque.4.Statistic methods: the data of germiculture is denoted by negative rate and do X2 test.Resultsl.The results of germiculture: The negative rate of the germiculture at the first week in No.l group is 60%, without notable difference with that in No.2 group (58.2%) but has a great difference with that in No.3 group (73.9%) and in No.4 group(zero).2.Results of vitreous check-up: The degree of vitreous opaque is approximately the same in No.l and No.2 group. The lowest degree is in No.3 group and the highest degree is in No.4 group.3.Results of pathologic check-up: there are about the same degree of retinal damage in No.l and No.2 group. The slightest damage is in No.3 group and the most serious damage is in No.4 group.DiscussionPurulent endophthalmitis can expand fast and damage eye tissue badly, if can't prevent and treat it in time and in effect, the result usually is to lose the vision even eyeball atrophy. So it is especially important to find it out early and adopt suited therapy.
