Research Into The Photodynamic Therapy Of Skin Hemangioma And It's Relationship With The Apoptosis Of Capillary Endothelium

[OBJECTIVE] Use the comb as skin hemangioma model, to investigate hemangioma histopathology change and the change of apoptosis of capillary endothelium after photodynamic therapy. Explore the mechanism of photodynamic therapy, afford experimental basis for photodynamic therapy of hemangioma.[METHODS] Use comb as animal model of skin hemangioma. There are six groups in this research. Group A (n=8) were normal comb; Group B (n=8) were treated by 10J/cm2of fluence of laser; Group C (n=8) were treated by 15J/cm2of fluence of laser; Group D (n=8) were just injected with HMME 10mg/kg; Group E (n=8) were injected with HMME 10mg/kg and then given 10J/cm2of fluence of laser; Group F (n=8) were injected with HMME 10mg/kg and then given 15J/cm2of fluence of laser. Samples of histology were collected respectively after 1st, 2nd , 3rd week after photodynamic therapy or laser therapy, Continually observe pathology change of capillary and the change of apoptosis of endothelium of capillary.[RESULT]: the ratio of reduction of capillary after laser therapy in group B is 48.58+5.10% and in group C is 57.58+8.17%,and the ratio of reduction of capillary after photodynamic therapy in group E is 61.26+16.87% and in group F is 68.56+10.54%。The reduction of capillary is more in group E and group F than it in group B and group C respectively (P<0.05). there are some fiber appear in the treated region in the 3rd week as well as the reduction of number and the diminution of diameter of capillary. There is no obviously proliferation of capillary with the come out of the apoptosis of some endothelium of capillary after photodynamic therapy or laser therapy. The effect of photodynamic therapy is better than that of laser therapy(P<0.05). [CONCLUSION]: It is effective in treating skin hemangioma using laser therapy or photodynamic therapy, the effect of photodynamic therapy is better than that of pure laser therapy. The appearance of apoptosis of endothelium of capillary indicate that one of the mechanism of laser therapy and photodynamic therapy maybe to induce apoptosis of endothelium of capillary.
