Objective:To investigate the association of PON1 Q191R gene polymorphism with type 2 diabetes mellitus accompanying hypertension and type 2 diabetes mellitus and essential hypertension and normal control subjects.Methods:1. Genomic DNA was extracted from peripheral blood leukocytes in 105 T2D patients, 82 T2D+EH patients, 90 EH patients and 58 control subjects. DNA was amplified by polymerase chain reaction (PCR). PCR product was digested with the restriction endonuclease ALWI and separated on 2% agarose gel electrophoresis.2. PON activity was estimated by photometer in 105 T2D patients, 82 T2D+EH patients, 90 EH patients and 58 control subjects.3. Serum immunoreactive insulin (IR1) and C-peptide (C-P) at fasting and oral glucose loading were measured by radio-immunoassay in 105 T2D patients, 82 T2D+EH patients, 90 EH patients and 58 control subjects.Results:1. The frequencies of QQ genotype and Q allele of T2D+EH patients were significantly higher than that of T2D patients (A^=3.84 T^O.05, X2=7.27 PO.05). There was no significant difference in frequency of Q allele among T2D patients and EH patients and control subjects (Jf2=0.041 />>0.05, ^=0.082 P>0.05, ^=0.223 P>0.05).2. PON activity was associated with PON genotype, serum PON activity increased in the order of QQ<0.01). PON activity of T2D patients was lower than that of EH patients and control subjects (F=7.38 P<0.01, F=8.24 (P<0.01), there was no significant difference between EH patients and control subjects(F=3.63 P>0.05).3. Logistic regression analysis results suggested that BMI and PON activity and PON genotype were the independent risk factors of diabetes mellitus accompanying hypertension.Conclusions:1. PON1 Q191R polymorphism was related to diabetes mellitus accompanying hypertension in Fujian province of China. QQ gene typeand Q allele were the independent risk factors of type 2 diabetes mellitus accompanying hypertension.2. PON activity was associated with PON genotype. QQ genotype and Q allele were associated with lower PON activity, however RR genotype and R allele were associated with higher PON activity.3. PON activity of T2D+EH patients was lower than that of T2D patients and EH patients and control subjects. In addition, PON activity of T2D patients was lower than that of EH patients and control subjects. These results suggested that PON gene polymorphism and PON activity were associated with the occurrence of type 2 diabetes mellitus accompanying hypertension.
