Paraoxonase-1 And Elderlg Patients With Type 2 Diabetes Mellitus

Objective:To study the relationship between serum paraoxonase-1 (PON-1) activity,Glu-Arg 192 polymorphism of PON-1gene in elderly patients with type 2 diabetes mellitus and diabetic complications.Methods: Serum PON-1activity was measured using phenylacetate as the substrate according to the method described by references in three weeks PON-1 genotype was determined by PCR-RFLP in 100 elderly cases with type 2 diabetes mellitus and 100 cases normal controls.Results: PON-1activity is much lower in 100 elderly eases with type 2 diabetes mellitus than that in 100 cases normal controls (92.07 + 33.02ku/L vs 132.29+ 19.74ku/L ,P<0.01) and PON-1activity is much lower in elderly patients with type 2 diabetic macroangiopathy (73.18 + 13.00ku/L vs 92.07 + 33.02 P<0.01) and PON-1activity is much lower in elderly patients with type 2 diabetic microangiopathy (74.33 +18.09ku/L vs 92.07+ 33.02ku/L,P<0.01). PON-1/HDL-C activity is much lower in elderly patients with type 2 diabetes mellitus and diabetic macroang-iopathy and diabetic microangiopathy (P<0.05 or P<0.01) The A.B allele frequencies of the PON-1gene are 0.44,0.56 and 0.39.0.61 in 100 cases normal controls and 100 elderly cases with type 2 diabetes mellitus respectively in the population of Chong qing .The A.B allele frequencies of the PON-1gene are 0.33.0.67 and 0.37.0.63 in elderly diabetic marcoangiopathy and diabetic microangiopathy.The genotype distribution of the PON-1polymorphism is not significantly different between the elderly patients with type 2 diabetes mellitus and diabetic vascular conplications and normal controls.Conclusions: Serum PON-1activity is much lower in elderly patients with type 2 diabetes mellitus and diabetic vascular complications.The lower serum PON-1activity may play-a role in elderly patients with type 2 diabetes mellitus and diabetic vasular complications. Glu-Arg 192 polymorphism of the PON-1gene is not associated with elderly patients with type 2 diabetes mellitus and diabetic vasular complication and the elderly population of Chong qing.
