Abstract/Summary:
The Purpose of this experiment is to study on effect of Astragalus Polysaccharids and Berberine on the laying performance, egg quality, serum biochemical indexes and intestinal enzyme activity, intestinal microflora on hen by extracting herbal medicine, experiment of feeding, slaughter test and sample analysis, providing theoretical basis for herbal medicine utilization on hens. The results are as follows:Test1:Astragalus Polysaccharids and Berberine were extracted respectively from Astragalus, and Berberis by water extraction. Astragalus Polysaccharids content was 50.52% and Berberine content was 16.40%,which were measured by colorimetry. 1200 Jianghan layers were divided randomly into 5 groups with 5 replicatesper grouP, 48 layers per replicate. The basic diet of the 5 groups was the same and treatments were as follow by order:100mg/kg APS+30mg/kg Ber, 200mg/kg APS+30mg/kg Ber, 100mg/kg APS+60mg/kg Ber, 200mg/kg APS+60mg/kg Ber. The results showed that every treatment laying rate were impeoved respectively 7.27%(P<0.01),6.30%(P<0.01),4.32%(P<0.05),2.69%(P>0.05),which were compared to control group.TreatmentⅠwas increased markedly with control group in daily egg quatity(P<0.05) and wasn,t discrepant with other treatments(P>0.05).On broken egg, treatmentⅡwas increaser significantly than treatmentⅢ(P<0.05). On sofe egg, compared with control group, TreatmentⅠ,ⅡandⅢwere reduced significantly(P<0.01); TreatmentⅢwere less decreased significantly than treatmentⅠ,Ⅱ, andⅣ(P<0.01).On matte egg, TreatmentⅣwere less decreased significantly than other treatments(P<0.01). On dead and bad hen, compared with control group, treatmentⅡwas reduced markedly (P<0.05), and other treatments didn't deffer(P>0.05.On daily feed intake, treatmentⅠ,Ⅱ, andⅣwere less decreased than control group(P<0.05), and every treatment wasn't differ. On average egg weight and deformed egg, every experimental group hadn't difference(P>0.05). In a short, adding APS and Ber on fodder could increase laying rate and average egg weight and decrease feeding intake.Test2:On the last day of the experiment, five eggs of per replicate were collected randomly for measureing egg's conventional and nutrious quanlity. The results showed that APS and Ber no effect on egg shape index, yolk index, yolk ratio, eggshell thickness, yolk color and moisture(P>0.05). Campared with control group, treatment group 2 is higher (P<0.01)on Haugh unit; treatment groups 1,2 and 4 decrease significantly(P<0.01) on Ether Extracts; treatment groups 1,2 and 3 increase significantly(P<0.01)on Crude protine; treatment grouPs 1and 4 are lower (P<0.01)on Cholesterol, treatment group 2 is low(P<0.05).Test3: On the last day of the experiment, one hen of per replicate were slaughtered randomly and collected blood. The results showed that total Protein, globulin and high density lipoprotein cholesterol of every treatment's serum were increased by effect of compound herbal medicine(P<0.05), and alkaline phosphatase of every treatment's serum were increased significantly by that(P<0.01)and low density liPoProtein cholesterol of every treatment's serum were decreased significantly by that. ( P<0.01 ) . There were increased significantly of treatmentⅡ' complement-4 and triglyceride(P<0.01), and decreased significantly of total cholesterol of treatmentⅠandⅢ(P<0.01).On LH and FSH of serum, Compared with control group, treatmentⅠwas increased significantly(P<0.01)and treatmentⅡwas improved(P<0.05).Test4: On the last day of the experiment, one hen of per replicate were slaughtered randomly and collected chime of small and the large intestine, whice were kePt in -80°C low temperature refrigerator. The results indicated that The contents of total protein, globulin, high-density lipoprotein cholesterol (HDL-C) and alkaline phosphatase (ALP) in all treatment groups were significantly (P < 0.05 or P < 0.01) increased comared with those in the control group. The cholesterol contents of all treatment groups were decreased significantly (P < 0.05 or P < 0.01) compared with those of the control group. The oPtimal traditional Chinese medicine compound preparation were the basic diets supplemented 200 mg / kg astragalus polysaccharides and 30 mg / kg berberine.
