Study On The Preparation Of The "CHAIQIN" Injection And Preliminary Pharmacodynamics

This study was for the common syndrome in the veterinary clinic with the main symptom of fever or alternate attack of chill and fever, accompanied by loss of appetite, vexation, nausea or vomiting and so on. From the view of traditional Chinese veterinary medicine theories, this syndrome could be classified into Shaoyang disease. So we studied on the classic prescription called "xiao-chai-hu-tang" used to treat Shaoyang disease for thousands of years through the observation of treatment to Shaoyang pathological model by its separated components. According to the results, we prepared the Chinese medicine injection of "Chaiqin", built its quality control method, and detected its treatment effect on the Shaoyang disease model. The results were as follows:1. Intraperitoneal injected of E. coli LPS (Lipopolysaccharides, LPS) with the dose of 40, 80,160μg/kg, the rats were showed obvious triphasic febrile response with good reproducibility and the saline-injected control group didn't exhibit obvious hyperthermia comparing with the LPS-injected rats. The group with 80μg/kg showed more obvious triphasic febrile response especially.2. We detected the function of immunity, gastrointestinal motility and liver and found the animals displayed obvious triphasic febrile phenomenon by 80ug/kg LPS ip had pathological changes in those systems indeed. The degraded gastrointestinal motility and increased aminotransferase (ALT), alkaline phosphatase (AKP), total bilirubin (TBil) and bile acid (TBA) in serum consistented with the description in "Shanghan Lun". With the temperature degraded, the WBC increased, which reflected the struggle of good and evil in Shaoyang disease to a certain extent. In the other hand, the typical prescription XCHT in traditional Chinese medicine was used to treat the disease, indicating that the XCHT had favorable effect on triphasic fever, not only relived fever, but also recuperated the disfunction of gastrointestinal motility and liver. The indexes didn't have significant difference with the control group except the TBil and TBA (P<0.05), and the number of lymphocytes and leukocytes were increased.3. Eight groups of separated components in xiao-chai-hu-tang were used to treat Shaoyang disease in rats. The results showed all the groups except the group with Bupleurum, Scutellaria, and ginger could obviously reduce the raised temperature, ALT and AKP, with no significant difference to the control group (P<0.05). That demonstrated the group with Bupleurum and Scutellaria had similar efficacy with xiao-chai-hu-tang on Shaoyang disease, so we chosen this group as our prescription for injection.4. Detected the baicalin and saikosaponin A in "chaiqin" Injection by high performance liquid chromatography (HPLC).The chromatographic condition of baicalin detection are as follows:Chromato bar:Hypersil ODS-C18,5μm,4.6×250mm; Detector:ultraviolet detector; Mobile phase:Methanol-aquaefer-orthophosphonic acid (45:55:0.2, V/V); Flow number:1mL/min; Column temperature:35℃; UV Detect W/L:278nm; Sample size: 10μL. Detect method:To measure sample solution 1milL exactly into measuring flask (100mL) and attenuate it to reach the graduate by mobile phase, misce bene, then sife it by filter membrane (0.45μm). Recipe 10μL for sample injection and HPLC detection, then record the peak area and caculate the content. The chromatographic condition of saikosaponin A detection are as follows:Chromato bar:Hypersil ODS-C18,5μm, 4.6×250mm; Detector:ultraviolet detector; Mobile phase:Methanol-aquaefer (75:25, V/V); Flow number:1mL/min; Column temperature:35℃; UV Det. W/L:208nm; Sample size: 10μL. Detect method:Sift the sample solution by filter membrane (0.45μm), recipe 10μL for sample injection and HPLC detection, then record the peak area and caculate the content. The methods could be used for the content detection for "chaiqin" Injection with the characteristic of highly stabilization, reliability, reproducibility, recovery rate and accurate consequence.5. The "chaiqin"Injection with the dose of 1.25mL/kg,0.625mL/kg and 0.313mL/kg were given to the rats with Shaoyang disease. The temperature, gastrointestinal motility, ALT and AKP had no significant difference with the control group (P<0.05), which demonstrated the "chaiqin" Injection had favorable effect on Shaoyang disease.
