| This subject studied on the effect of three medicines (exogenous nitric oxide, praziguantel embrocation and xiaochaihutang) respectively using to cure Schistosomiasis japanum or eliminate liver granuloma causing by the same illness. The results show that the optimized ratio among NO liposome materials (water, cane sugar, Tween-80 and beeswax) is 15:2.0:2.3:1.0. The best absorbing way is oil phase absorbing first, which is the mixing phase between beeswax and Tween80, and water phase absorbing between water and cane sugar followed. The particle of liposome made in this way has relatively smaller diameter, better stability and higher efficiency in absorbing NO. So it can be as exogenous NO to cure Schistosomiasis japanum on mice. The result shows that exogenous NO liposome not only has some medical efficiency on Schistosomiasis japanum (the worm-reduction rate obtains 45%, and the egg-reduction rate obtains 42.7%), but also can obtain the efficiency as liver targeting drug releasing. In addition, several solubility curves of different impregnates were investigated, and the two solvents (1, 4-dioxane and ethylene glycol monophenyl ether (EPH)) that can better dissolve praziguantel were obtained, and they were also made into embrocation to carry out the experiments on rabbits. According to the experiment about two solvents, praziguantel has the best solubility in ethylene glycol phenyl ether (49.1% was obtained at 35 centigrade degree). However, the effect of penetration is bad and even added penetration enhancers (PE) did not improve the efficiency of penetration. Besides, praziguantel has the relatively higher solubility in 1, 4-dioxane (48.8% was obtained at 45 centigrade degree). As a penetration embrocation, the penetration efficiency is very good even without penetration enhancers. And using it as an embrocation, the experiment therapy on mice artificially infected by Schistosomiasis cercariae reveals that both the worm-reduction rate and egg-reduction rate are 100%. Furthermore, Xiao Chai Hu Tang(XCHT), which was made from decoction, distillation and concentration of Radix Buplerui, Rhizoma Pinelliae, Radix Seutel-lariae, Fructus Ziziphi, Radix Ginseng, Radix Glycyrrhizae and Rhizoma Zingiberis Recens, has better effect on the liver granuloma causing by Schistosomiasis japanum which came from intragastrical administration therapy to Kunming mice, and 58.8% of egg-reduction rate was obtained. After donoring the liver in the mouse which takes XCHT group as medicine, we found that the color of liver became lighter and the number of eggs in liver reduced from 473 to 195 by the electron microscopy. To some extent XCHT can eliminated part of the egg node. |
