In order to understand the cycle variety discipline of the different stages dairy goats ovarian corpus luteum organizational structure,to definite the corpus luteum composition cell type,the structure characteristics of each type and the variety discipline of different periods cells. The experimental material is estrous cycle dairy goats in this study, using conventional materials histologic paraffin - HE dyeing methods and transmission electron microscope techniques keep under observation each period structure of goat ovarian. Using immunohistochemistry SP method to study the positioning and variation distribution of AR and P450arom in the estrous cycle and pregnancy dairy goats ovarian, combine with ovarian endocrine physiological characteristics,to research the function and significance of AR and P450arom in the female reproductive and function of ovarian. Results as follows:1. The surface of goat corpus luteum is membrane,made of big lutein cells(granulosa lutein cells) and small lutein cells(theca lutein cells),the big lutein cells is more than small lutein cells. mainly located in the central part of the corpus luteum. small lutein cells in the surrounding of corpus luteum and a part of between the the big lutein cells. The big lutein cells can be divided into two,the two the big lutein cells is different from the form, the dyeing,and the nuclear shape. The morphological structure of corpus luteum cells is different in different stages of cycle. Through the cytoplasm of colouring in big lutein cells and the vary of secretory granule under electron microscopy display that secretory activity will gradually declined with processes of lifecycles, collagen fiber of lutein cells is collection bundles. Results show that the structure organization of lutein cells is change in different stages of cycle,the degradation lutein cells are increase with processes of lifecycles;The different estrous cycle stage goats ovarian corpus luteum configuration can also affect the number of growth follicles and atresia follicles.2. The AR and P450arom positive outcome is mainly located in the membrane progesterone, smooth the outer membrane and lutein cells, progesterone and stromal cells. Reaction products mainly located in forming and cytoplasmic, from quantity and colouring strength of positive cells and positive gray value analysis,the express of two kinds protein in estrous cycle strong than early and late estrus cycle. 3. Immune positive response of the AR and P450arom the general tendency is more to less, colouring parts transfer membrane to the cytoplasm.the average gray value of different phase of pregnancy is highly significant(P<0.01). In the early pregnancy theca cells and granular layer have weak positive reaction,but after middle stage the weak positive reaction only happen to theca. Follicles granule cells and big lutein cells both have P450 positive reaction in pregnancy goat ovarian, the positive reaction of lutein cells is gradually decreasing with the process of pregnancy,althougth individual lutein cells have expressed the enhancement in late pragnancy,but but overall intensity of reaction are relatively weak.The expression discipline of Two proteins in estrous cycle and pregnancy is on speaking terms with animal reproductive endocrine characteristics in different stages,point out that AR and P450arom can participate in different periods adjustment of reproductive physiology ovarian.
