| Jining gray goat is a species of lamb leathers in China, characterized by precosity and high productivity. The initial mating of female goats happened approximately 6 months old. The average yean percentage of primiparity is 163.1, general speaking, one year two fetus or two years three fetus, average yean rate is 293.7%. Luteinizing hormone is the important reproductive hormone, which specially bind with the corresponding receptors play it's physiological function. Luteinizing hormone receptor not only exists in the gonads, as ovary and testis, but also exists in the uterus and fallopian tube and the other gential tract of the femail mammal. To invest the high productivity mechanism of goat, the study Using GAPDH as an inner control, Real Time Fluorescence Quantitative RT-PCR method was established to analyze the levels of LHR mRNA expression; Applied the method of immunohistochemistry to study the distribution and localization of LH receptor in the different segment of the oviduct of four estrous cycle phases in Jining Gray Goat. The main result is as follows:1. LHR mRNA expression was different in the oviduct of Jining gray goat during the four estrous cycle phrases. The LHR mRNA relative expression was different in the infundibulopelvic of faliopian tube during the four estrous cycle phrases, being highest in the estrus, lowest in the diestrus, intermediate in the proestrus and the metestrus. The relative expression in the estrus was very significantly higher than in other oestrous cycle phases (P<0.01). The LHR mRNA relative expression was different in the ampolla of the oviduct during the four estrous cycle phrases, being highest in the proestrus, lowest in the metestrus, intermediate in the estrus and the diestrus. The relative expression in the metestrus was significantly lower than in other oestrous cycle phases (P<0.05). The LHR mRNA relative expression was different in the isthmus of the oviduct during the four estrous cycle phrases, being highest in the proestrus, lowest in the metestrus, intermediate in the estrus and the diestrus. The relative expression in the proestrus was very significantly highest than in other oestrous cycle phases (P<0.01).2. The localizations of LHR in the infundibulum, ampolla and isthuus of the oviduct of proestrus, estrus, metestrus and diestrus were carried out in Jining Gray Goat respectively. The results showed that LHR-immunoreactive was distributed over the oviduct during the four estrous cycle phrases, and it was mainly distributed over the epithelium of tubal mucosa, lamina propria, and blood vessel endothelium. The presence of LHR in many tubal cell types could mean that LH directly regulates tubal functions such as transport, maturation, and fertilization of gametes, early embryonic development, and transport and synthesis of specific glycoproteins, independent of ovarian steroid hormones. |
