| Yimeng black goat is a choiceness breed of which its both of meat, cashmere and skin could be used. At present, the study on the gonadotropin receptor (FSHR and LHR) are mainly focused on testis and ovary, FSH stimulate the development and maturation of ovarian follicles as well as the production of estrogen in granulosa cells, and affect the function of Sertoli cells and thus indirectly affect sperm formation. LH was mainly direct the production of androgen in theca cells,which provide substrate for the synthesis of estrogen in granulosa cells, and initiate ovulation and maintain the production of progesterone in corpus luteum, and stimulate androgen production of Ledig cells in testis, which provide appropriate environment for the development of sperm. Gonadotropin binds its specific receptor in the membrane of target cells, which lead to a conformational change of the ligand-receptor complex, furthermore, regulate the activity of adenylate cyclase, and then catalyse adenosine triphosphate (ATP) into cyclic adenosine triphosphate (cAMP), which activates cAMP-dependent protein kinase, leading to the synthesis of the corresponding mRNA and a new effective protein. Untill now, the FSHR gene and LHR gene expressing in the nongonadal system of goat rarely reported. To investigate the variation law of mRNA and protein expression levels of FSHR gene and LHR gene in uterus of low productive Goats, total RNA was extracted from the uterus of YiMeng Black goat and Real Time Fluorescence Quantitative PCR method (real-time qRT-PCR) was established so as to analyse mRNA levels of FSHR gene and LHR gene in uterus of four estrous cycle phases in YiMeng Black Goat. Applied the method of immunohist- ochemistry to study the distribution and localization of FSH receptor and LH receptor in the different segment of the uterus of four estrous cycle phases in YiMeng Black Goat. The main result is as follows: 1. mRNA of FSHR gene and LHR gene were expressed in the uterus during the four estrous cycle phrases. The expression of FSHmRNA was higher than that of LHRmRNA. In cornua uteri, the expression of FSHmRNA was highest in metestrus and lowest in proestrus; in uteri caruncle the expression of FSHmRNA was highest in diestrus and lowest in proestrus and there was no obvious difference among the four phases(P>0.05). In cervical, the expression of FSHmRNA was highest in estrus and lowest in diestrus, which is significantly different with other phases(P<0.05). In cornua uteri, uteri caruncle and cervical, the expression of LHRmRNA was highest in diestrus, which is significantly different with the rest phases(P<0.05). In uterine body, the expression of FSHmRNA and LHRmRNA were low and no obvious law was observed.2. Immunopositive staining cells was found in the uterine horn and uterine caruncle endometrial luminal epithelial cells. In the lamina propria with scattered immunopositive reaction; in the uterine glands of glandular cells and endothelial cells revealed no positive staining; in uterus and cervix in the glandular epithelial cells and luminal epithelial cells show negative reaction and so on; FSHR during the estrous cycle of other weak positive reaction. In diestrus and uterine caruncle uterine horn endometrial luminal epithelial cells, showing strong positive reaction LHR; in the lamina propria, muscle layer and serosal layer were not found to have positive; uterine body and cervix of the same negative reaction; estrous cycle Other less positive during the LHR. |
