Follicle-stimulating hormone (FSH) is a glycoprotein hormone secreted by the anterior pituitary,which plays an important role in the gonadal development and the reproductive modulation.The physiological function of FSH is mediated by the specific receptor of the granulosa cells--follicle stimulating hormone recepter(FSHR). FSHR was found successively in the follicle of the ovary, the oocyte,the granulosa cells, the oviduct, and the uterus, after the first discovery in the surface of sertoli cells by Means in 1972. FSHR has functional roles in promoting differentiation of the theca interna cells, the proliferation of the granulosa cells, and it also stimulates development, enlargement and maturity of the follicle. Moreover, there is a close relationship between the expression level of FSHR mRNA in ovary and reproduction rate. Therefore,study on the expression characteristics of FSHR mRNA in ovarian tissue of Jining Grey goats plays an important role in exploring the mechanism of FSHR, accelerating follicular growth and differentiation, facilitating the formation of the dominant follicles and improving the reproductive rate of the domestic animals which has low fecundity.Immunohistochemistry and Real Time Fluorescence Quantitative PCR method were used to analyze the location of FSHR and the levels of FSHR mRNA gene in the ovary of four estrous cycle phases in Jining Gray Goat. What the results shows as follows,1.A certain regularity of the expression characteristics of FSHR mRNA was showed during the ovary of four estrous cycle phases, highest in estrous and lowest in metestrous. The expression level increased gradually in diestrous, proestrus, estrus,metestrus.2.The results of the gene expression showed that the ratio of the expression level of FSHR mRNA in ovary during estrous cycle is metestrus (0.3500±0.1316)0.05).3.The FSHR positive cells in ovarian tissue of Jining Grey goats was showed in granulosa cells,theca cells in follicle. The results indicates that the quantity of the FSHR positive cells increased with the growth level of follicle. FSHR was not found in corpus luteum during metestrus.4. The quantity of FSHR positive cell in different estrous cycle stage is as follows: estrus>proestrus>diestrous>metestrus.
