| This paper mainly studied the dynamic changes of Th1/Th2 cytokines and biochemical indexes in the estrous cycle,early pregnancy,before and after abortion on goats,analyzed change law of cytokines and biochemical indexes to research its physiological significance and provide theoretical basis for increasing the reproductive efficiency and embryo transplantation pregnancy rate of goats.25 clinical health white goat ewes were selected for the test animals,natural mating after the third estrous,21 of pregnancy.At day 1,2,3,7,10,14,17,19 of estrous cycle and at day 0,1,3,5,8,11,15,18,20, 26,30,40,50,60 of pregnancy collected blood from jugular veins and separated serum.At day 1,7,14,19 of estrous cycle collected uterine fluid,diluted with physiological saline to 5 times,then after centrifugation separated supernatant.The contents of TNF-α,IL-2,IL-10 were detected by ELISA and the contents of NO,Ca,Pi,AKP,GPT and UN in serum were detected with biochemistry method.The results showed that in the estrous cycle,from day 1 to 14 the contents of TNF-αin turn increased,at days 17,19 gradually decreased.The contents of IL-2 from day 1 to 10 continued to increase,at day 14 started to decrease and at day 19 improved.The contents of IL-10 from day 1 to 3 gradually decreased,at day 7 increased,then decreased again,at day 17 down to the lowest point,at day 19 improved.In the beginning of estrous at day 1-3 NO increased,then decreased and at day 14 increased again;In early pregnancy the contents of TNF-αand IL-2 in serum increased at first and then decreased,the contents of IL-10 continued to increase.In early pregnancy NO gradually increased,TNF-αand IL-2 in serum had a positively correlated trend and a negatively correlated trend with IL-10.NO had a negatively correlated in early pregnancy with TNF-αand IL-2,had a positively correlated with IL-10;the contents of biochemical indexes(NO except)in estrous cycle nearly kept constant;in early pregnancy,from day 0 to 60 the contents of serum Ca and UN decreased gradually.From pregnancy day 0 to 60 the contents of serum Pi,AKP and GPT increased slowly.In the estrous cycle the contents of TNF-α,IL-2 and IL-10 in serum were very significantly lower than in uterine fluid(P<0.01),cytokines had a similar trend reaching significant correlation.The contents of cytokines in serum of goats could indirectly reflected the trend of expression Level of cytokines in uterine fluid.8 goats were randomly selected from 21 pregnancy ones,at day 60 of pregnancy neck muscles were injected PG(0.2mg/goats),after 24h were injected PG 0.1mg/goats again.The results showed that in the second injection PG 12-24h,all 8 goats were abortion,the abortion rate was 100%.Before injection PG and at day 1,3,8,10 of abortion collected blood and separated serum.Through the testing of samples before and after the abortion(as above),the contents of TNF-αafter abortion were very significantly higher than those before abortion(P<0.01),at day 1,3 of abortion increased gradually,at day 8,10 decreased gradually.The contents of IL-2 after abortion were significantly higher than those before abortion(P<0.05),at day 1,3,8 of abortion increased gradually,at day 10 slightly decreased.The contents of IL-10 after abortion were significantly lower than those before abortion(P<0.05 or P<0.01),from abortion day 1 to 10 decreased systematically.The contents of NO after abortion were significantly higher than those before abortion(P<0.05),at day 1,3 of abortion increased gradually,at day 8,10 decreased.Except at day 1 of abortion,at day 3,8,10 the contents of Ca were higher than those before abortion,but the difference was not significant(P>0.05),from abortion day 1 to 10 increased gradually.The contents of Pi after abortion were significantly lower than those before abortion(P<0.05),from abortion day 1 to 10 increased gradually.The contents of serum AKP after abortion were very significantly lower than those before abortion(P<0.01),from abortion day 1 to 10 decreased gradually.The contents of serum GPT after abortion were lower than those before abortion,the difference was significant or not(P<0.05 or P>0.05),from abortion day 1 to 10 decreased gradually.the contents of UN after abortion were significantly higher than those before abortion(P<0.05),from abortion day 1 to 10 increased gradually. |
