Research On Quality And Fingerprint Of Folium Eucommia Of ChongQing

Eucommia,(Eucommia ulmoides Oliver),which has functions of detoxification,anti-bacterial, anti-inflammatory,choleretic and significant immunity enhancement,has a good prospect.It has been used for thousands of years.Eucommia is used for tradition.The research showed that Folium Eucommia had the same chemical composition and effects with Eucommia essentially,even some certain compents in Folium Eucommia were higher than that of in Eucommia.Folium Eucommia had been indexed by《Chinese Pharmacopoeia》(2005 Edition),and make chlorogenic acid as the component indicator of the quality control.ChongQing is one of the main origin of Eucommia.In order to take advantage of this Chinese herbal and to control its quality,we do researches on the quality of Folium Eucommia produced in ChongQing systematically.This content mainly include:1 Pharmacognosy identification of Folium Eucommia produced in ChongQingWe have done reaearches on the pharmacognosy identification by observing the outward appearance,microstructure and the TLC identification of Folium Eucommia produced in ChongQing.Results:The outward appearances of Folium Eucommia produced in ChongQing showed no differences;The microstructure of different ages of Folium Eucommia were similar.The TLC had found that the samples had the same colour fluorescent spots.It showed that the outward appearance of Folium Eucommia had little to do with the origin,the microstructure of different ages of Folium Eucommia had little difference,and TLC identification declared the authenticity of Folium Eucommia produced in ChongQing.According to the《Chinese Pharmacopoeia》(2005 Edition),we did the following examinations, moisture,total ash,acid insoluble ash and ethanol-soluble extract.Finally,the results all coincided with the standards.It pointed that the quality of Folium Eucommia produced in ChongQing was stable.2 Determination of heavy metals of Folium Eucommia produced in ChongQingDetermination of Pb,Cd,As of Folium Eucommia produced in ChongQing were studied by atomic absorption spectrometry and atomic fluorescence spectrometry.Results showed that the Pb and Cd contents conformed with the standards,but the As contents of Folium Eucommia produced in some areas were excessive.So,the contents of As of Folium Eucommia produced in ChongQing should be further monitored.3 Determination of chlorogenic acid of Folium Eucommia produced in ChongQing Firstly,the different extraction solvents and ways were studied to observe the effects on chlorogenic acid extraction rate.Orthogonal design was used to optimize the extraction of chlorogenic acid process. Then,the chlorogenic acid in Folium Eucommia and Eucommia were measured by HPLC.As a result, the content of chlorogenic acid of Folium Eucommia of different areas of ChongQing had difference, and the contents of the samples from NanChuan and ChengKou were higher.The study also found that content of chlorogenic acid of Folium Eucommia was negatively correlated with ages.Besides,the content of chlorogenic acid in Folium Eucommia was much higher than that of in Eucommia.The content of Chlorogenic acid of Folium Eucommia harvested at different time had shown a sinigificant difference(P＜0.01),the samples harvested in June were the highest.We can see from the above results ChengKou should be the dominated planting areas.Folium Eucommia has medicinal effectiveness indeedly,it should be well developed.Taking advantage of Folium Eucommia saplings can be a new way of producing chlorogenic acid.Harvest time has great influence to the content of chlorogenic acid and june is the best.4 Establishment of HPLC fingerprint ofFolium Eucommia produced in ChongQingWith the method of HPLC,we obtained 18 mutual characteristic peaks by analyzing these chromatograph peaks of 10 batchs of samples,selected 13 strong peaks as the characteristic fingerprint peak,and analyzed relative peak area and relative retention time of these 13 strong peaks to obtain the digital characteristic HPLC-fingerprint of Folium Eucommia produced in ChongQing.This digital characteristic HPLC-fingerprint can control the quality of Folium Eucommia produced in ChongQing.5 Establishment of quality standards of Folium Eucommia produced in ChongQing.Based on the above-mentioned studies,the quality standards of Folium Eucommia produced in ChongQing have been established preliminary,which provide basic datas for the quality control of F olium Eucommia produced in ChongQing.
