Effective chemical ingredients are the basis for disease prevention and treatment using herbs. Since these ingredients are stored in particular tissue and cell structures, it is of great significance in theory and practice to determine the tissue structures of medicinal parts of medicinal plants and study the relationship between the medicinal ingredients and accumulation thereof and the structure and development of medicinal plants by using histochemical methods.The paper takes Apocyni Veneti Folium as the subject of research, and carries instrumental analysis in regards to the development of its tissue structure, hostochemical localization of effective ingredients, and main effective ingredients in leaves of Apocynum venetom L. (Apocyni Veneti Folium) harvested from different sites and at different harvest time, aiming to illustrate the correlation between the development process of the structure of Apocyni Veneti Folium and dynamic accumulation of effective ingredients, so as to provide an assessment index for selective growth of high-quality Apocyni Veneti Folium and provide a scientific basis for proper harvesting of Apocyni Veneti Folium grown in Jiangsu. The research mainly includes the following aspects.1. Literature study. Literatures related to Apocyni Veneti up to now are studied thoroughly.2. Anatomical study on the structural development of Apocyni Veneti Folium. By using a traditional paraffin section method, anatomical microscopic observation is carried out on leaves at different sites, the internal structural change during the development of leaves is studied, and microscopic measurement is carried out on diachyma tissues of Apocyni Veneti Folium at different stages of growth. The conclusion indicates that Apocyni Veneti Folium has typical characteristics of xeromorphism leaves.3. The histochemical localization of main chemical composition in Apocyni Veneti Folium. By using the method of simple histochemical and fluorescent staining, to determine the site of accumulation of flavones and to interpret the regular pattern for flavones distribution of leaves at different harvest time, microscopic inspection is carried out. Cellular and organizational localization and relative quantification of flavones has been fulfilled by the technology of laser cofocal scanning. The organizational localization of flavones about the different mother nucleus has been done. The localization of tannins and naphtha has been done by simple histochemical. Result show that flavones distributes in epidermis cell, cuticle, collenchyma, vessel. The more distribution is at upper epidermis. The accumulation is also at diachyma in mature leavrs. Tannins distributes in diachyma, phloem, laticifer. Naphtha accumulates in palisade tissue.4. The content determination of main chemical composition in Apocyni Veneti Folium in different growth period and different site. The content analysis of epigallocatechin, rutin, hyperin, isoquercitrin, astragalin and volatile ingredient in Apocyni Veneti Folium in different growth period and different site is carried out by HPLC and HSGC-MS. Result show that the optimum harvest season is bud period. The content of astragalin showed the decreased regulation at the leaves of Apocynum venetom L. of the different site. The contents of hyperin and isoquercitrin were higher at June and July. The content of volatile ingredient is keeping rising trend at June July and August.5. the analysis of Apocyni Veneti Folium in different growth period and commodity medicinal materials by UPLC-ESIMSn. The qualitative and quantitative analysis of the main flavones in Apocyni Veneti Folium in different growth period and commodity medicinal materials has been done by UPLC-ESIMSn. The comparison between the leaf of A.hendersonii and A. venetom has been done particularly. The conclusion illustrate that the content of kaempferol glycosides show rising trend at upper to lower leaves in Apocyni Veneti Folium. To distinguish the leaf of A.hendersonii and A. venetom, the former is characterized by Quercitin-3-O-maltoside, the latter is characterized by hyperin. The content difference has exist between the leaf and flower of A. venetom L..Innovations and Features:1. It is the first time to systematically analyze the structural development of Apocyni Veneti Folium, and illustrate the structural characteristics of Apocyni Veneti Folium at different periods of growth.2. Hostochemical localization is carried out on main flavones compounds of Apocyni Veneti Folium under a laser cofocal microscope by using an induced fluorescent staining method, and the localization may reach the cell level. Hostochemical localization is carried on different mother nucleus flavones compounds based on different light emitting spectra of the main of flavones compounds after fluorescence induction. Also, the distributions of flavones compounds in Apocyni Veneti Folium at different periods of growth are also analyzed and compared, based on which an accumulation rule and influencing factors of flavones compounds in Apocyni Veneti Folium are proposed.3. It is the first time to compare the contents of main flavones compounds in Apocyni Veneti Folium at different periods of growth and different sites. Also, the main chemical ingredients of the leaves of A.hendersonii and A. venetom are compared.
